TGF-β1诱导的Postn、α-SMA和Collagen Ⅰ在宫腔粘连内膜组织中表达上调且与粘连程度相关

目的:骨膜蛋白(Postn)参与多种纤维化疾病的病理形成,且与促纤维化因子TGF-β1的信号通路活动密切相关,但在宫腔粘连(IUAs)形成过程中的作用机制尚不清楚。本研究通过检测不同程度IUAs患者子宫内膜组织中Postn与纤维标志物(α-SMA,CollagenⅠ)的mRNA和蛋白质表达,并从细胞水平研究TGF-β1作用与三种目标蛋白质表达,从而探讨Postn、α-SMA和CollagenⅠ在IUAs发病过程中的作用机制,及与IUAs严重程度间的关系。方法:收集明确诊断的轻、中和重度IUAs患者内膜组织各20例(实验组),非宫腔粘连者20例(对照组),利用实时荧光定量PCR技术、Western印迹技术和免疫组化染色分别检测Postn、α-SMA和CollagenⅠ在不同粘连程度IUAs患者的子宫内膜组织中mRNA和蛋白质表达差异。同时,Western印迹技术分别检测原代ESCs经TGF-β1溶液不同时间、不同浓度作用后Postn、α-SMA和CollagenⅠ蛋白表达情况。结果:Postn与α-SMA、CollagenⅠ在IUAs患者子宫内膜组织中的mRNA和蛋白质表达水平趋势一致,...
