肿节风联合番茄红素对内毒素致急性肺损伤SD大鼠炎症反应的影响

【摘要】:目的:探讨肿节风注射液与番茄红素联合使用对ALI大鼠肺损伤炎症水平的影响。 方法:健康雄性250~280gSD大鼠,50只,均分成5组(随机),经实验分组处理后再通过腹腔用注射器注入6mg/kg的脂多糖建立ALI模型: NC组(n=10):一般对照组(NS处理); LPS组(模型组,n=10),只给与LPS建模,不给药处理; LPS+肿节风组(S组,n=10):连续七天腹腔注射肿节风注射液(2.5g/kg); LPS+番茄红素组(L组,n=10):连续2周番茄红素油树脂灌胃(5mg/kg,每天两次); LPS+肿节风+番茄红素组(U组,n=10):连续两周番茄红素油树脂(5mg/kg,每天两次)、连续七天(第2周开始)腹腔注射肿节风注射液(2.5g/kg); 分组干预后第二天建模,建模后2h(T1)、6h(T2)和10h(T3)采集静脉血,最后一次采血后取大鼠肺组织标本,称重,取右肺中叶,做病理HE切片,分离血清,采用ELISA法检测TNF-α和IL-8含量的变化;计算各组大鼠肺指数,应用光学显微镜观察肺组织病理HE结果,分级大鼠肺泡炎并计分,统计分析实验结果。 结果: 1.肉眼观察:NC组大鼠肺脏外观正常,肺包膜无肿胀,颜色淡红,标本表面没有看到点状或片状出血。LPS模型组大鼠肺脏充血比较明显、标本体积肿大,能发现出血点或出血斑;L组、S组、U组大鼠肺脏标本炎症充血情况好于模型组。 2.肺指数的比较:LPS组大鼠肺指数明显高于其他四个组(P0.01)其余各干预组大鼠肺指数也有不同程度的增高,但均较LPS组增幅小;L组、S组、U组与NC组四组之间的肺指数差异不大。 3.肺组织HE染色比较:NC组大鼠肺组织无或偶见少量炎症细胞浸润;LPS组大鼠肺泡之间的间距变得更宽,数量众多的各类炎性细胞浸润在肺间质,肺泡腔隙被挤压变窄、有的腔隙内可见出血和(或)渗出物;较LPS组,L组、S组和U组炎性渗出情况均有不同程度的减轻,而U组减轻明显。 4.肺泡炎分级比较:LPS组肺泡炎评分均显著高于其他组;L组、S组和U组肺泡炎评分小于LPS组(P0.05或P0.01),而大于NC组(P0.05)。L组、S组和U组比较相差不明显,U组在干预组(L组、S组、U组)内的肺泡炎评分最低。 5.血清中TNF-α含量变化比较:T1时间点LPS组大鼠血清中TNF-α含量明显高于NC组、L组、S组和U组,此差异在统计学上有意义(P0.05或P0.01),L组和S组TNF-α含量较NC组均有升高,此差异在统计学上有意义(P0.05),U组TNF-α含量较NC组有升高,但无统计学意义,各干预组之间比较无统计学意义。T2时间点LPS组大鼠血清中TNF-α含量比NC组、L组、S组和U组仍增高,在有统计学上有意义(P0.05),S组和L组TNF-α含量较NC组稍有升高,无统计学意义,U组TNF-α含量较NC组稍有降低,各干预组之间比较无统计学意义,,T3时间点LPS组大鼠血清中TNF-α含量仍高于NC组、L组、S组和U组,此差异在统计学上有意义(P0.05),但NC组、L组、S组和U组之间比较无统计学意义。随着实验时间延长,LPS组、L组和S组TNF-α含量逐渐降低。 6.血清中细胞因子IL-8含量变化比较:T1时间点LPS组大鼠血清中IL-8含量高于NC组、L组、S组和U组,但比较在统计学上无意义,L组、S组和U组IL-8含量大致与NC组持平。T2时间点LPS组大鼠血清中IL-8含量明显高于NC组、L组、S组和U组,此差异统计学上有意义(P0.05或P0.01),L组IL-8含量比NC组升高,此差异统计学上有意义(P0.05),S组和U组TNF-α含量较NC组有升高,此差异未见统计学意义,各干预组之间比较统计学上无意义,T3时间点LPS组大鼠血清中IL-8含量高于NC组、L组、S组和U组,该差异统计学上有意义(P0.05),其余四组之间比较统计学上无意义。随着实验时间延长,LPS组和各干预组(L组、S组和U组)IL-8含量先升高后降低。 结论:肿节风注射液联合番茄红素能减轻内毒素血症所致的肺脏炎症反应,能在一定程度上对ALI时大鼠的受损肺脏予以保护。
[Abstract]:Objective: To investigate the effect of Zhongjiefeng injection and lycopene combined effect on inflammatory lung injury of ALI rats.
Methods: 50 healthy male 250~280gSD rats were divided into 5 groups (random). After the experimental group was processed, the ALI model was established by injecting 6mg/kg lipopolysaccharide into the abdominal cavity with injector.
Group NC (n=10): general control group (NS treatment);
Group LPS (model group, n=10) was only modeled with LPS, and no drug treatment was given.
LPS+ group (group S, n=10 zjf): seven consecutive days intraperitoneal injection of zjf injection (2.5g/kg);
LPS+ LYC group (group L, n=10) for 2 weeks lycopeneoleoresin gavage (5mg/kg, two times a day);
LPS+ + zjf lycopene group (group U, n=10) for two weeks lycopeneoleoresin (5mg/kg, two times a day), for seven consecutive days (second weeks) intraperitoneal injection of zjf injection (2.5g/kg);
The intervention group second days after modeling, after modeling 2H (T1), 6h (T2) and 10h (T3) in venous blood, blood for the last time after taking the sample, lung tissues of rats weighing, take the right middle lobe, pathological HE slice, separation of serum were measured by ELISA TNF- alpha and IL-8 content the rats in each group; lung index, lung tissue HE results observed by light microscope, grading of alveolitis in rats and scoring, analysis of the experimental results of statistics.
