迷走神经电刺激对脑外伤后昏迷大鼠前额叶皮层Orexin-A及受体OX1R表达变化的影响

【摘要】:目的:探讨迷走神经电刺激(Vagus nerve stimulation,VNS)对脑外伤(Traumatic brain injury,TBI)昏迷大鼠的促醒效果及Orexins在上述效应中的调节机制。方法:将120只SD大鼠随机分为四组:空白对照组、TBI组、刺激组(TBI+VNS)和拮抗剂组(TBI+VNS+SB334867)。通过VNS刺激脑外伤后昏迷大鼠,通过意识状态六级评分法评估大鼠的行为学变化,并于刺激结束后6h、12h、24h,用ELISA、Western-blot、免疫组织化学技术检测各组大鼠前额叶皮质Orexin-A和OX1R的表达。结果:TBI组中8只(8/30)大鼠出现翻正反射,刺激组中20只(20/30)大鼠出现翻正反射,拮抗剂组中12只(12/30)大鼠出现翻正反射;Orexin-A的含量在各时间点中呈现:拮抗剂组空白对照组TBI组刺激组(P0.05),而OX1R的表达在各时间点缺乏一致性,6h:空白对照组拮抗剂组TBI组刺激组(P0.05);12h:空白对照组TBI拮抗剂组刺激组(P0.05);24h:拮抗剂组空白对照组TBI组刺激组(P0.05)。结论:VNS可改善脑外伤昏迷大鼠意识状态水平,其作用机制可能与上调前额叶皮层中Orexin-A和OX1R表达水平有关。
[Abstract]:Objective: to investigate the effects of vagus nerve stimulation (VNSs) on the wake-up of rats with traumatic brain injury-induced brain injury (TBI) and the regulatory mechanism of Orexins in the above effects. Methods: 120 Sprague-Dawley rats were randomly divided into four groups: the blank control group, the control group, and the control group. The rats in the stimulation group and the antagonist group were treated with TBI VNS SB334867. The behavioral changes of the rats were evaluated by the six grade score of consciousness state after VNS stimulation. The expressions of Orexin-A and OX1R in the prefrontal cortex of rats in each group were detected with ELISAA Western-blot and immunohistochemistry. Results the positive inversion reflex was found in 8 / 30 of the rats and 20 / 30 / 30 of the rats in the stimulation group. In the antagonist group, the content of Orexin-A appeared in 12 / 30 rats at different time points: in the antagonist group, the level of Orexin-A was found in the blank control group, the stimulation group in the TBI group was P0.05A, and the expression of OX1R was lack of consistency at each time point for 6 h: TBI in the blank control group of antagonist group. The control group was treated with TBI antagonist group for 24 hours. The control group with TBI group was stimulated with P0.05. Conclusion the level of conscious state of coma rats with brain injury can be improved by using the control group (P0.05) for 12 h, and the control group with TBI antagonist group for 24 h, and the control group, TBI group, can improve the level of consciousness state of the rats with brain injury. The mechanism may be related to the up-regulation of Orexin-A and OX1R expression in prefrontal cortex.
