大鼠创面愈合的非线性光谱成像研究

【摘要】:目的:在本研究中,我们运用基于双光子激发荧光(TPEF)和二次谐波产生(SHG)的非线性光谱成像技术对创伤愈合过程中胶原蛋白等物质进行持续监测,从而初步明确其在创面愈合过程中的动态变化规律,指导临床对创伤愈合过程的干预。方法:选用雄性SD大鼠90只,建立大鼠背部脊柱两侧圆形皮肤全层切除创伤模型,脊柱右侧创面用“外用重组牛碱性成纤维细胞生长因子”(Recombinant Bovine Basic Fibroblast Growth Factor For External Use贝复济)进行干预,设为实验组;脊柱左侧创面应用生理盐水,设为对照组,进行局部用药,同体对照,记录创面愈合时间,测量创面面积并计算创面愈合率。分别于造模后的1、2、3、5、7、10、14、20d,随机选取10只作为活体扫描标本,利用双光子显微镜成像系统活体动态监测创面胶原蛋白的SHG成像信息,结果:实验组与对照组在造模后17天时的创面愈合率分别为100%和95.32%,实验组创面愈合时间明显缩短。实验及数据分析结果清晰的显示了大鼠皮肤创面愈合过程中胶原蛋白等物质的动态变化,在创面愈合过程中,胶原蛋白呈明显的向心性生长。同一时间,实验组较对照组胶原蛋白的合成分泌有显著增多,,统计学差异明显。结论:非线性光谱成像技术是基于双光子激发荧光(Two-PhotonExcitation Fluorescence TPEF)和二次谐波产生(Second Harmonic GenerationSHG)的生物医学光学领域的新技术,以其简单、无创且可在体成像等特点,在将来有望成为继病理切片之后的另一种组织形态学研究乃至临床诊断的利器。随着科技进步及光谱成像设备的小型化发展,非线性光谱成像技术在基础研究与临床应用中拥有巨大的发展潜力及广阔的应用前景。
[Abstract]:Objective: in this study, we used nonlinear spectral imaging based on two-photon excited fluorescence (TPEF) and second harmonic generation (SHG) to continuously monitor collagen and other substances during wound healing. Therefore, the dynamic changes of the wound healing process were preliminarily clarified, and the clinical intervention on wound healing was guided. Methods: 90 male SD rats were selected to establish the model of full-thickness skin excision on both sides of the dorsal spine of rats. The right spine wound was treated with "recombinant Bovine Basic Fibroblast Growth Factor For External Use" as experimental group, and the left spine wound was treated with normal saline as control group. In the same body control, the wound healing time was recorded, the wound area was measured and the wound healing rate was calculated. Two-photon microscope imaging system was used to monitor the SHG imaging information of collagen in vivo. Results: the wound healing rates of the experimental group and the control group were 100% and 95.32, respectively, and the healing time of the experimental group was significantly shortened. The results of experiment and data analysis clearly showed that the glue was used in the healing process of the rat skin wound. The dynamic changes of proteins and other substances, At the same time, the synthesis and secretion of collagen in the experimental group was significantly higher than that in the control group. Conclusion: nonlinear spectral imaging is a new technique in biomedical optics based on Two-Photon Excitation Fluorescence (TPEF) and second harmonic generation (second Harmonic Generation SHG), which is simple, noninvasive and in vivo imaging. In the future, it is expected to be another kind of sharp weapon for histomorphology research and clinical diagnosis after pathological section. With the development of science and technology and the miniaturization of spectral imaging equipment, Nonlinear spectral imaging technology has great development potential and broad application prospect in basic research and clinical application.
