HBV相关ACLF患者PBMCs中ROG、GATA3和T-bet mRNA水平检测

【摘要】:目的检测HBV相关慢加急性肝衰竭(ACLF)患者外周血单个核细胞(PBMC)中GATA3抑制因子(ROG)、GATA3和T-bet mRNA水平变化及意义。方法收集45例HBV相关ACLF(HBV-ACLF)患者和15例健康志愿者的外周血,分离单个核细胞,提取总RNA,用实时定量PCR的方法检测ROG、GATA3和T-bet mRNA水平。结果 HBV-ACLF患者和健康对照ROG水平分别为0.98±0.27和0.64±0.19,差异有统计学意义(P0.05);HBV-ACLF患者和健康对照GATA3水平分别为0.73±0.15和0.58±0.22,差异有统计学意义(P0.05);HBV-ACLF患者和健康对照T-bet水平分别为0.78±0.29和0.46±0.17,差异有统计学意义(P0.05);HBV-ACLF患者和健康对照ROG/GATA3和T-bet/GATA3比较,差异均有统计学意义(P0.05)。HBVACLF患者ROG表达水平与GATA3 mRNA呈负相关(r=-0.724,P0.05)。而ROG mRNA表达水平与T-bet/GATA3比值无相关性(r=-0.064,P0.05)。结论 ROG、GATA3和T-bet在HBV-ACLF患者PBMC中表达水平上调明显,参与肝功能衰竭的发病发生和发展。
[Abstract]:Objective to investigate the changes and significance of GATA3 inhibitory factor (GATA3) in peripheral blood mononuclear cells (PBMCs) of patients with HBV associated with chronic and acute hepatic failure (ACLF).Methods Peripheral blood samples were collected from 45 patients with HBV associated with HBV-ACLF and 15 healthy volunteers. Mononuclear cells were isolated, total RNAs were extracted, and the levels of ROGN GATA3 and T-bet mRNA were detected by real-time quantitative PCR.There were significant differences in ROG/GATA3 and T-bet/GATA3 between patients with HBV-ACLF and healthy controls.There was a negative correlation between the expression of ROG and GATA3 mRNA in patients with P0.05. HBV-ACLF, and there was a negative correlation between the expression of ROG and GATA3 mRNA (P 0.05).However, there was no correlation between the expression of ROG mRNA and the ratio of T-bet/GATA3.Conclusion the expression of Gata _ 3 and T-bet in PBMC of HBV-ACLF patients is up-regulated, which is involved in the pathogenesis and development of liver failure.
