脑损伤与他克莫司促进周围神经再生的对比研究

【摘要】:目的通过比较脑损伤、他克莫司促进大鼠坐骨神经再生修复的机制,探索脑损伤促进周围神经损伤修复机制。方法实验动物随机分为4组,颅脑损伤合并坐骨神经损伤模型(A组),单纯坐骨神经损伤模型(B组),造模后分别给予A1组他克莫司、A2组等量生理盐水、B1组他克莫司、B2组等量生理盐水,2周。分别于术后第4、8、12周,进行测定坐骨神经指数、腓肠肌恢复率、肌肉Masson染色、神经组织苏木素-伊红染色、Masson染色、甲苯胺蓝染色,术后8、12周测定坐骨神经干的动作电位幅值恢复率、辣根过氧化物示踪检测。结果通过实验比较,脑损伤联合应用他克莫司促进周围神经损伤的修复效果坐骨神经指数、神经干的动作电位幅值恢复率、腓肠肌恢复率、肌肉Masson染色、神经组织苏木素-伊红染色、Masson染色、甲苯胺蓝染色、辣根过氧化物示踪检测优于其它单独因素的作用。结论脑损伤与他克莫司促进周围神经损伤的修复的机制不同,其促进周围神经损伤修复的机制仍需进一步探讨。
[Abstract]:Objective to compare the mechanism of brain injury and tacrolimus promoting sciatic nerve regeneration and repair in rats and explore the mechanism of brain injury promoting peripheral nerve injury. Methods the experimental animals were randomly divided into 4 groups. The model of craniocerebral injury combined with sciatic nerve injury (group A) and the model of simple sciatic nerve injury (group B) were treated with tacrolimide A2 group (A1 group) and tacrolimus B2 group (B 1 group) respectively for 2 weeks. The sciatic nerve index, gastrocnemius recovery rate, muscle Masson staining, hematoxylin-eosin staining and toluidine blue staining were measured at the 8th week after operation. The recovery rate of action potential amplitude of sciatic nerve trunk and horseradish peroxide tracer were measured 812 weeks after operation. Results the effects of tacrolimus combined with brain injury on the repair of peripheral nerve injury were compared. The sciatic nerve index, the recovery rate of action potential amplitude of nerve trunk, the recovery rate of gastrocnemius muscle and Masson staining of muscle were observed. The effects of hematoxylin-eosin staining Masson staining toluidine blue staining horseradish peroxide tracer detection were superior to those of other single factors. Conclusion the mechanism of brain injury is different from that of tacrolimus in promoting the repair of peripheral nerve injury.
