乌司他丁联合亚低温对心肺复苏术后大鼠的脑保护

【摘要】:目的:构建心脏骤停-心肺复苏成功的大鼠模型,进一步研究亚低温和乌司他丁联合治疗对心肺复苏大鼠脑组织的保护作用。 方法:采用窒息法制备大鼠的心肺复苏模型。随机分成五组:假手术组、对照组、亚低温组、乌司他丁药物组、乌司他丁联合亚低温组,治疗结束后于1、3、6小时各取血液2ml,通过ELISA分别检测各组血液中炎症细胞因子TNF-α、IL-6、IL-8的表达。手术6小时后处死大鼠,取脑组织,光镜检测脑组织病理形态变化。 结果:(1)血清中炎症因子表达量:与对照组比较,亚低温组、乌司他丁组、联合组在短时间内(3小时内)均可抑制IL-6,IL-8,TNF-α的表达且效果显著(P0.05),联合组的抑制效果更为明显。当时间延长到6小时,联合组抑制效果与亚低温组或乌司他丁组差异无统计学意义(P0.05)。(2)脑组织病理形态变化:心肺复苏后各组均有不同程度的脑部损伤,亚低温组、乌司他丁组、脑部损伤程度有所减缓,联合组损伤程度最轻。 结论:乌司他丁和亚低温的联合应用可抑制心肺复苏术后大鼠的炎症反应,减缓脑损伤。图6幅,表4个,参考文献65篇
[Abstract]:Aim: to establish a successful rat model of cardiac arrest and cardiopulmonary resuscitation (CPR) and to study the protective effects of mild hypothermia and ulinastatin on brain tissue of cardiopulmonary resuscitation (CPR) rats. Methods: the rat cardiopulmonary resuscitation model was established by asphyxia. They were randomly divided into five groups: sham operation group, control group, mild hypothermia group, ulinastatin drug group, ulinastatin combined with mild hypothermia group. The blood samples were collected at 1, 3 and 6 hours after treatment. The expression of inflammatory cytokines TNF- 伪 and IL-6,IL-8 in the blood of each group were detected by ELISA. The rats were killed 6 hours after operation, the brain tissue was taken and the pathological changes of brain tissue were detected by light microscope. Results: (1) the expression of inflammatory factors in serum: compared with the control group, the mild hypothermia group, ulinastatin group and combined group could inhibit the expression of IL-6,IL-8,TNF- 伪 in a short time (3 hours) and the effect was significant (P0.05), the inhibitory effect of the combined group was more obvious than that of the control group. When the time was prolonged to 6 hours, there was no significant difference in the inhibitory effect between the combined group and mild hypothermia group or ulinastatin group (P0.05). (2). After cardiopulmonary resuscitation, there were different degrees of brain injury in the combined group, mild hypothermia group and ulinastatin group. The degree of brain injury was alleviated, and that of the combined group was the least. Conclusion: the combination of ulinastatin and mild hypothermia can inhibit the inflammatory response of rats after cardiopulmonary resuscitation and slow down brain injury. 6 figures, 4 tables, 65 references
