低功率氦氖激光对小鼠烫伤创面血管内皮细胞因子和细胞凋亡因子基因表达的影响

【摘要】:目的:观察低功率氦氖激光照射后烫伤小鼠创面的变化及其对小鼠创面炎症介导因子VEGF、Caspase-3基因表达的影响。方法:60只烫伤小鼠模型随机分为激光照射组与对照组,其中激光照射组行低强度氦氖激光照射烫伤创面,能量密度15J/cm2,对照组采用假照射,能量密度0J/cm2,每天一次,每次15min,连续14d。分别在烫伤后0、1、3、7、14d记录创面面积,计算创面愈合率,并取全层创面组织标本,采用RT-PCR法检测VEGF、Caspase-3的基因表达。结果:激光照射组小鼠创面愈合率明显高于对照组(P0.05)。与对照组相比,激光照射组VEGF基因表达在烫伤后1d时降低,3d时开始增加(P0.05),7d时达到高峰(P0.01),随后降低,在14d时,VEGF基因表达已低于对照组(P0.05)。激光组1d时Caspase-3基因表达减少(P0.05),3d时Caspase-3基因表达持续降低(P0.01),7d时开始增高(P0.05)。结论:低功率氦氖激光照射可以加速烫伤创面愈合,可能与其调节VEGF和Caspase-3因子表达相关。
[Abstract]:Aim: to observe the effect of low power He-Ne laser irradiation on burn wound and the expression of inflammatory mediating factor (VEGF,Caspase-3) in burn mice. Methods: sixty scalded mice were randomly divided into laser irradiation group and control group. The laser irradiation group received low intensity He-Ne laser irradiation on the burn wound, the energy density was 15J / cm ~ 2, while the control group was treated with false irradiation, the energy density was 0 J / cm ~ 2, once a day. Each time 15 minutes, continuous 14 days. The wound area was recorded and the rate of wound healing was calculated on the 14th day after scald, and the VEGF,Caspase-3 gene expression was detected by RT-PCR method. Results: the wound healing rate in laser irradiation group was significantly higher than that in control group (P 0.05). Compared with the control group, the expression of VEGF gene in laser irradiation group decreased at 1 day after scalding, began to increase at 3 days (P0.05), reached its peak at 7 days (P0.01), and then decreased. At 14 days, the expression of VEGF gene was lower than that in control group (P0.05). In the laser group, the expression of Caspase-3 gene decreased at day 1 (P0.05), the expression of Caspase-3 gene continued to decrease at day 3 (P0.01), and the expression began to increase at day 7 (P0.05). Conclusion: low power He-Ne laser irradiation can accelerate wound healing, which may be related to the regulation of VEGF and Caspase-3 expression.
