壳聚糖蜂蜡膏对大鼠创面中CD34及羟脯氨酸的影响

【摘要】:目的观察壳聚糖蜂蜡膏对大鼠创面中CD34、羟脯氨酸的影响及创面愈合效果并探讨其可能机制。 方法取20只雄性SD大鼠,在其背部两侧制造全层皮肤缺损创面,制成创面模型,每只大鼠背部三个创面随机分为实验组、对照组和空白对照组。造模后每隔2d换药1次,空白对照组不做处理,实验组创面使用5g壳聚糖蜂蜡膏,对照组使用5ml生理盐水。免疫组化法评价创面组织中CD34表达情况,计数创面中新生毛细血管数量;酸水解法检测羟脯氨酸在创面组织中的表达;观察创面的未愈合面积、创面愈合率;计量资料用均数±标准差(X±s)表示,采用SPSS13.0统计学软件。两样本均数比较应用配对t检验,率的比较应用卡方检验。P0.05为差异具有统计学意义。 结果实验组第7、14、21天时创面组织中CD34表达量高于同期对照组;第7天时实验组毛细血管计数高于对照组,但无统计学差异(P0.05),第14、21天时实验组毛细血管数均高于同期对照组(表1),有统计学差异(P0.05)。实验组第7、14、21天时创面组织中羟脯氨酸含量均高于同期对照组(表2),差异有统计学意义(P0.05)。第7天实验组创面愈合率大于对照组,但无统计学差异(P0.05);第14d、21d实验组创面愈合率高于同期对照组(表3),具有统计学意义(P0.05)。 结论壳聚糖蜂蜡膏能促进创面组织中CD34、羟脯氨酸表达及毛细血管再生,可能是促进创面愈合的机制之一。
[Abstract]:Objective to observe the effect of chitosan beeswax ointment on CD34, hydroxyproline in rat wound and to explore its possible mechanism. Methods 20 male SD rats were used to make full-thickness skin defect wounds on both sides of their backs. Each rat was randomly divided into experimental group, control group and blank control group. Every 2 days, the blank control group was not treated, the experimental group was treated with 5 g chitosan beeswax ointment, and the control group was treated with 5ml saline. Immunohistochemical method was used to evaluate the expression of CD34 in wound tissue and count the number of neocapillaries in the wound, acid hydrolysis method was used to detect the expression of hydroxyproline in the wound tissue, and the unhealed area and wound healing rate of the wound were observed. The measurement data were expressed by mean 卤standard deviation (X 卤s), and SPSS13.0 software was used. The average of two samples was compared by paired t test, and the rate was compared by chi-square test. P0.05 was statistically significant. Results the expression of CD34 in the wound tissue of the experimental group was higher than that of the control group at day 7 and day 14. On the 7th day, the capillary count in the experimental group was higher than that in the control group, but there was no statistical difference (P0.05). On the 14th day, the capillary count in the experimental group was higher than that in the control group (table 1), and there was statistical difference (P0.05). The content of hydroxyproline in the wound tissue of the experimental group was significantly higher than that of the control group (P 0.05). On the 7th day, the wound healing rate in the experimental group was higher than that in the control group, but there was no statistical difference (P0.05); on the 14th day, the wound healing rate in the experimental group was higher than that in the control group (table 3), which had statistical significance (P0.05). Conclusion chitosan beeswax ointment can promote the expression of CD34, hydroxyproline and capillary regeneration in wound tissue, which may be one of the mechanisms of promoting wound healing.
