人膝关节滑膜细胞的增殖、分泌和趋化的体外研究

【摘要】:第一部分人膝关节滑膜细胞的体外培养 目的:获得人滑膜B型细胞,进行后续研究。方法:提取滑膜组织,消化,培养,传代,鉴定。结果:获得人滑膜B型细胞。结论:本方法可以获得人滑膜B型细胞,满足实验的需求。 第二部分tTG. β-NGF及bFGF对人膝关节滑膜细胞的作用 目的:研究tTG、βNGF及bFGF对人膝关节滑膜B型细胞的作用。方法:将第一部分培养好的滑膜细胞铺板,加入不同浓度的tTG、β-NGF及bFGF,抽取上清,测Ⅰ型胶原蛋白的含量,加入MTT,测细胞增殖,利用Transwel l测趋化作用。结果:tTG在20ng/ml之前,对滑膜B型细胞有促进增殖、分泌作用,之后,随着浓度的增加,抑制细胞增殖和分泌Ⅰ型胶原蛋白,tTG对滑膜B型细胞无趋化作用。β-NGF对滑膜B型细胞有抑制增殖和分泌Ⅰ型胶原蛋白作用,β-NGF对滑膜B型细胞无趋化作用。bFGF对滑膜B型细胞有促进增殖、分泌作用,bFGF对滑膜B型细胞无趋化作用。结论:在滑膜B型细胞增殖和分泌Ⅰ型胶原蛋白方面,tTG有双重作用,β-NGF有抑制作用,bFGF有促进作用;在滑膜B型细胞趋化方面,三种均无影响。
