阿尔茨海默病患者外周血淋巴细胞周期分布及p27表达的改变及意义

【摘要】:目的研究阿尔茨海默病(Alzheimer’s disease,AD)患者外周血淋巴细胞细胞周期分布及周期素依赖性蛋白激酶抑制剂p27(p27)、细胞周期素E(cyclin E)表达的改变及意义。方法选取40例AD患者和30例正常对照者外周血,立即用密度梯度离心法分离淋巴细胞,用流式细胞分析仪检测淋巴细胞细胞周期分布,分析DNA相对含量(DNA relative contents,DNARC)和细胞增殖指数(cell proliferation index,PI),用免疫印迹法检测淋巴细胞p27、cyclin E的表达水平。结果与正常对照组相比,AD组外周血淋巴细胞分布于G1期的比例(G1%)显著下降而S%显著升高(P0.01);淋巴细胞DNARC和PI显著升高(P0.01);p27在AD组表达显著下降(P0.01)。轻、中重度AD组之间外周血淋巴细胞周期的改变及p27水平无统计学差异(P0.05)。结论 AD早期外周血淋巴细胞即显示了增强的增殖能力和细胞周期调节的紊乱,且与病情进展无关,淋巴细胞p27水平有可能成为AD早期诊断的外周标志物。
[Abstract]:Objective to study the cell cycle distribution of peripheral blood lymphocytes and the expression of cyclin-dependent protein kinase inhibitor p27p27, cyclin E(cyclin in patients with Alzheimer's disease (AD). Methods 40 patients with AD and 30 patients with AD were enrolled in this study. Peripheral blood of normal controls, Lymphocytes were separated by density gradient centrifugation and the cell cycle distribution was detected by flow cytometry. The relative content of DNA relative contentsDNA) and cell proliferation index (Pi) were analyzed. The expression of p27 cyclin E in lymphocytes was detected by Western blotting. Results compared with the control group, the percentage of peripheral blood lymphocytes in G 1 phase in AD group was significantly higher than that in control group (P < 0.05). The expression of DNARC and Pi in lymphocytes decreased significantly in AD group and decreased significantly in AD group. There was no significant difference in the changes of lymphocyte cycle and the level of p27 between moderate and severe AD patients. Conclusion Peripheral blood lymphocytes in the early stage of AD showed increased proliferative ability and disorder of cell cycle regulation, which was not related to the progression of the disease. The level of lymphocyte p27 may be a peripheral marker for early diagnosis of AD.
