IL-1、IL-18、IL-33基因多态性与迟发型阿尔茨海默病的相关性研究

【摘要】:目的:探讨炎症因子IL-1α基因(rs1800587)、IL-1β基因(rs1143627)、IL-18基因(rs187238)和IL-33基因(rs11792633)单核苷酸多态性(SNP)位点与湖南汉族人群迟发型阿尔茨海默病(LOAD)的关系。 方法:运用病例-对照研究,选取LOAD患者201例为AD组,另选择年龄及性别相匹配的健康人群257例为对照组。利用聚合酶链反应(PCR)和基质辅助激光解吸电离飞行时间质谱技术(MALDI-TOF-MS)对炎症因子IL-lα、IL-1β、IL-18、IL-33共四个SNPs位点(rs1800587、rs1143627、rs187238和rs11792633)进行基因分型检测,比较不同基因型、等位基因频率在两组间的差别,并分析四个SNPs位点与LOAD的相关性。 结果: 1.IL-1α基因rs1800587各基因型频率在两组间差异无统计学意义(P0.05),CC型和(CT+TT)型及C、T等位基因频率在两组间差异有统计学意义(P0.05),携带T等位基因者具有较高的LOAD发病风险(OR=1.592,95%CI=1.037-2.443)。 2.IL-1β基因rs1143627及IL-18基因rs187238各基因型及等位基因频率在两组间差异无统计学意义(P0.05)。 3.IL-33基因rs11792633各基因型及等位基因频率在两组间差异有统计学意义(P0.05),携带T等位基因者具有较低的LOAD发病风险(OR=0.648,95%CI=0.498-0.843)。 4.多因素非条件Logistic回归分析示文化程度、丧偶、总胆固醇、IL-1α基因rs1800587及IL-33基因rs11792633与LOAD显著相关。 结论: 1.IL-1α基因rs1800587位点多态性与湖南汉族人群LOAD易感性相关联,T等位基因可能是湖南汉族人群LOAD的危险因素。 2.IL-1β基因rs1143627位点多态性和IL-18基因rs187238位点多态性与湖南汉族人群LOAD易感性无关联。 3.IL-33基因rs11792633位点多态性与湖南汉族人群LOAD易感性相关联,T等位基因可能是湖南汉族人群LOAD的保护因素。
[Abstract]:Objective: to investigate the relationship between the single nucleotide polymorphisms (SNPs) of the inflammatory factor IL-1 伪 gene rs1800587 and IL-33 gene rs11792633) and the late onset Alzheimer's disease (Lod) in Hunan Han population. Methods: a case-control study was conducted in which 201 patients with LOAD were selected as AD group and 257 healthy subjects matched with age and sex as control group. Polymerase chain reaction (PCR) and matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) were used to detect four SNPs loci (rs1800587rs1143627rs187238 and rs11792633) of inflammatory factor IL-l 伪, IL-1 尾 and IL-18IL-33. The correlation between four SNPs loci and LOAD was analyzed. Results: There was no significant difference in the frequency of rs1800587 genotypes of 1.IL-1 伪 gene between the two groups. There was no significant difference between the two groups in the frequency of rs1800587 genotypes (P0.05C and TTCT) and the frequencies of CT-alleles were significantly different between the two groups. The patients with T alleles had a higher risk of LOAD. There was no significant difference in the frequency of 2.IL-1 尾 rs1143627 and IL-18 rs187238 genotypes and alleles between the two groups (P 0.05). The frequencies of rs11792633 genotypes and alleles of 3.IL-33 gene were significantly different between the two groups (P 0.05). Those with T allele had a lower risk of LOAD. 4. Multivariate conditional Logistic regression analysis showed that education level, widowed degree, total cholesterol IL-1 伪 gene rs1800587 and IL-33 gene rs11792633 were significantly correlated with LOAD. Conclusion: The rs1800587 polymorphism of 1.IL-1 伪 gene is associated with the susceptibility to LOAD in Hunan Han population. The T allele may be the risk factor of LOAD in Hunan Han population. The polymorphism of 2.IL-1 尾 gene rs1143627 locus and IL-18 gene rs187238 locus were not associated with LOAD susceptibility in Hunan Han population. The polymorphism of 3.IL-33 gene rs11792633 locus is associated with the susceptibility to LOAD in Hunan Han population. The T allele may be the protective factor of LOAD in Hunan Han population.
