雷帕霉素对β淀粉样蛋白25-35诱导PC12细胞损伤的保护作用

【摘要】:目的雷帕霉素可通过提高自噬水平,改善阿尔茨海默病特征性病理,但其内在机制尚需进一步研究。文中旨在探讨雷帕霉素对β淀粉样蛋白25-35(Aβ25-35)诱导PC12细胞损伤的保护作用机制。方法取对数生长期细胞PC12分为5组:对照组(等量无血清DMEM)、模型组及10μmol/L雷帕霉素组、40μmol/L雷帕霉素组、160μmol/L雷帕霉素组(分别用加入10、40、160μmol/L的雷帕霉素),除对照组外,其余各组均用20μmol/L的Aβ25-35作用于PC12细胞构建细胞损伤模型。结果 (1)与模型组细胞存活率[(51.47±2.59)%]、凋亡率[(52.22±2.33)%]比较,10、40、160μmol/L雷帕霉素组、对照组细胞存活率[(54.64±2.42)、(64.79±2.91)、(56.50±2.55)、(99.98±0.73)%]明显升高,凋亡率[(45.33±2.83)、(36.89±2.85)、(48.00±2.83)、(3.33±2.45)%]明显降低(P0.05)。(2)模型组p-PKB表达量为0.33±0.01,p-m TOR表达量为1.97±0.05,p-tau表达量为2.09±0.19;与模型组上述指标比较,10、40、160μmol/L雷帕霉素组、对照组p-PKB表达量(0.37±0.01、0.42±0.01、0.40±0.01、0.44±0.02)明显升高(P0.05),而p-m TOR表达量(1.64±0.05、0.66±0.04、0.35±0.01、0.62±0.01)和p-tau表达量(2.02±0.15、1.79±0.05、1.86±0.06、1.53±0.04)明显降低(P0.05)。结论雷帕霉素能提高Aβ25-35损伤的PC12细胞的存活率,降低细胞凋亡率,对Aβ25-35致PC12细胞损伤有保护作用。其机制可能与雷帕霉素抑制p-m TOR,负反馈调节PI3K/PKB/m TOR信号转导通路,减少tau蛋白过度磷酸化有关。
[Abstract]:Objective rapamycin can improve the characteristic pathology of Alzheimer's disease by increasing autophagy, but its intrinsic mechanism needs further study. The aim of this study was to investigate the protective mechanism of rapamycin on 尾 -amyloid 25-35 (A 尾 25-35) -induced injury in PC12 cells. Methods PC12 cells in logarithmic growth phase were divided into 5 groups: control group (equal dose without serum DMEM), model group and 10 渭 mol / L rapamycin group (40 渭 mol / L rapamycin group, 160 渭 mol / L rapamycin group). The other groups were treated with A 尾 25-35 at 20 渭 mol / L for PC12 cells.
