【摘要】:目的以施万细胞作为周围神经模型,比较富血小板纤维蛋白(PRF)与浓缩生长因子(CGF)对其的影响。方法随机选取18~55岁身体健康志愿者10人,无菌条件下采集静脉血10 m L,制备PRF和CGF。将细胞随机分为对照组、PRF组和CGF组。倒置相差显微镜下观察细胞形态;MTT比色法测定细胞增殖能力;酶联免疫吸附法检测细胞上清液中神经生长因子的分泌量;流式细胞仪检测细胞周期。结果倒置相差显微镜下观察各组细胞形态无明显差异;MTT结果显示PRF组和CGF组与对照组相比光密度值明显升高(P0.05),细胞上清液中神经生长因子分泌量均明显升高(P0.05),处于S+G2M期的细胞数明显增多(P0.01),而PRF组与CGF组相比各项指标的差异无统计学意义。结论 PRF和CGF均可以促进施万细胞增殖并增加神经生长因子的分泌量,但就改善细胞增殖能力、促进神经生长因子分泌来看,二者无明显差异。
[Abstract]:Objective to use Schwann cells as peripheral nerve model. The effects of platelet rich fibrin (PRF) and concentrated growth factor (CGF) on it were compared. Methods 10 healthy volunteers aged from 18 to 55 years were randomly selected. PRF and CGF were prepared by collecting 10 mL venous blood under aseptic condition. The cells were randomly divided into two groups: control group and CGF group. The morphology of the cells was observed under inverted phase contrast microscope. Cell proliferation was measured by MTT colorimetry. Enzyme linked immunosorbent assay (Elisa) was used to detect the secretion of nerve growth factor (NGF) in the supernatant. Results there was no significant difference in cell morphology under inverted phase contrast microscope. MTT results showed that the optical density of PRF group and CGF group was significantly higher than that of control group, and the secretion of nerve growth factor in cell supernatant was significantly higher than that of control group (P 0.05). The number of cells in S G 2m phase increased significantly (P 0.01). Conclusion both PRF and CGF can promote the proliferation of Schwann cells and increase the secretion of nerve growth factor. However, there was no significant difference between the two groups in terms of improving cell proliferation and promoting the secretion of nerve growth factor (NGF).
