近β钛合金TLM表面双层辉光离子渗氮改性对成骨细胞早期功能的影响

【摘要】:目的:观察近β钛合金Ti-3Zr-2Sn-3Mo-25Nb(TLM)双层辉光离子渗氮改性层表面成骨细胞早期黏附、增殖、碱性磷酸酶活性、成骨相关基因的表达情况,研究TLM渗氮改性层对成骨细胞早期功能的影响。方法:近β钛合金TLM圆片试样经抛光、清洗后平均分成两组,实验组试样采用双层辉光离子渗氮处理,TLM抛光组作为对照。将小鼠颅顶前骨细胞亚克隆14接种于两组试样表面,扫描电镜观察接种4h后细胞的黏附形貌;MTT法测3d、5d、7d细胞增殖情况;检测7d、14d细胞碱性磷酸酶(alkaline phosphatase,ALP)活性;实时荧光定量聚合酶链反应(quantitative real time,qRT-PCR)检测细胞Runt相关转录因子2(Runt-related transcription factor-2,RUNX2)、Ⅰ型胶原α1链(typeⅠcollagen alpha 1 chain,COLⅠα1)、骨保护素(osteoprotegerin,OPG)和核因子-κB受体活化因子配体(receptor activator of nuclear factor-kappaB ligand,RANKL)mRNA的表达情况。结果:接种4h,TLM渗氮改性组细胞黏附形态良好,有细丝状伪足伸出,细胞间连接紧密;培养3d,实验组细胞量(0.277±0.007)明显高于对照组(0.249±0.004)(P=0.004)而培养5d和7d两组细胞量均未见明显差异;培养14d,实验组细胞ALP活性(173.606±1.884)明显高于对照组(162.582±2.426)(P=0.003);渗氮组RUNX2、Ⅰ型胶原α1链、骨保护素mRNA相对表达量均显著高于对照组(P0.05),RANKLmRNA相对表达量显著低于对照组(P=0.016)。结论:近β钛合金TLM表面采用双层辉光离子渗氮改性后,改性层促进成骨细胞早期黏附,利于成骨细胞分化,提高细胞骨保护素OPGmRNA表达量,同时抑制了RANKLmRNA的表达。推测在改性层表面可能更快速形成骨结合。
[Abstract]:Aim: to observe the early adhesion, proliferation, alkaline phosphatase activity and osteoblast-related gene expression of the surface osteoblasts of Ti-3Zr-2Sn-3Mo-25NbTLM layer modified by double glow plasma nitriding. The effects of TLM nitriding modified layer on the early function of osteoblasts were studied. Methods: after polishing and cleaning, the samples of near 尾 -Ti alloy TLM wafer were divided into two groups. The experimental group was treated with double glow ion nitriding and treated with TLM as control group. The subclone 14 of mouse precranial bone cells was inoculated on the surface of the two groups. The cell adhesion morphology was observed by scanning electron microscope (SEM) and MTT method was used to measure the proliferation of the cells for 3 days and 5 days after inoculation. Alkaline phosphatase (ALP) activity was detected on day 14 after 7 days.
