烟草浸提液对人牙周膜细胞HSP70表达的影响

【摘要】：目的:观察烟草浸提液(ST)对人牙周膜细胞(hPDLCs)增殖、热休克蛋白70(HSP70)表达的影响。方法:常规培养hPDLCs,第5~8代细胞用于实验。对照组为常规培养液,实验组为含不同浓度的ST培养液(0.016~50 g/L ST),作用24 h后MTT法检测细胞增殖、免疫组化和Western Blot法检测细胞HSP70的表达。结果:ST浓度为0.4~50 g/L时抑制hPDLCs增殖,增强细胞质及胞核内HSP70的表达;ST浓度增加,其作用更明显。结论:ST可抑制hPDLCs增殖,促进HSP70呈表达。
[Abstract]:Objective: to observe the effect of tobacco extract (STS) on the proliferation and expression of heat shock protein 70 (HSP70) in human periodontal ligament cells. The experimental group was treated with different concentrations of ST-containing 0.016 ~ 50 g / L STN. The proliferation of hPDLCs was detected by MTT method after 24 h exposure, and the expression of HSP70 was detected by immunohistochemistry and Western Blot method. Results the proliferation of hPDLCs was inhibited when the concentration of St was 0.4g / L, and the expression of HSP70 was detected by Western Blot method. The increased expression of HSP70 in cytoplasm and nucleus increased the concentration of St, and the effect was more obvious. Conclusion: 1. Conclusion: 1. St can inhibit the proliferation of hPDLCs and promote the expression of HSP70.
