不同时间、不同力值条件下牙周组织内白介素-17表达变化的研究

【摘要】:目的 用SD大鼠建立正畸牙根吸收模型,探讨白介素-17在不同作用力及不同加力时间条件下牙根吸收区的表达变化。 方法 选择SD雄性成年大鼠,要求月龄相同、体重相近,建立正畸牙根吸收模型,按不同正畸作用力分为伪实验组、40g、60g、80g三个组,每组根据加力时间分为1d、4d、7d、14d四个亚组。采用连续切片行苏木精-伊红染色、抗酒石酸酸性磷酸酶染色观察牙根吸收情况。应用免疫组织化学方法及双抗夹心酶联免疫吸附法(ELISA)观察白介素-17的表达变化情况。 结果 1)牙根吸收区域主要表达在根中1/3,相同力值不同加力时间的情况下,牙根吸收在加力第4d时已经发生,到第7d逐渐达到峰值,第14d牙根吸收程度逐渐下降,牙本质修复现象在部分根吸收陷窝出现;加力时间相同不同力值的情况下,随着正畸力值的增加,,牙根吸收的程度不断加重; 2)伪实验组未见白介素-17阳性表达。相同力值不同加力时间条件下,表现为第1d主要表达为弱阳性,随时间增加,第7d时达到峰值,表现为强阳性,随后至第14d,表达强度逐渐下降,表现为中度阳性。相同时间力值越大,阳性表达强度越明显。白介素-17的表达与加力时间和力值之间的相关性具有统计学意义(P0.05),且与力值大小呈正相关; 3)牙根吸收面积在不同加力时间和力值大小条件下,与白介素-17阳性表达率呈正相关(P0.05),有统计学意义。 结论 1)牙根吸收程度在不同加力时间及力值大小时存在规律性,力值越大,牙根吸收越严重;在一个加力周期内,牙根吸收程度从第4d到第7d呈现上升趋势,7d时达到峰值,随后逐渐下降,第14d牙根吸收区出现牙本质修复现象; 2)白介素-17量的变化与牙根吸收变化趋势相同。正畸牙周组织改建过程中有白介素-17参与,提示临床操作中应注意加力大小和周期的控制; 3)白介素-17可能参与正畸牙根吸收过程。
[Abstract]:Purpose. A orthodontic root resorption model was established in SD rats to investigate the expression of interleukin-17 (IL-17) in the root resorption region under different applied forces and time. Method. Adult SD male rats were selected to establish the orthodontic root resorption model with the same age and similar body weight. According to different orthodontic forces, the rats were divided into three groups: the pseudo-experimental group (40g / 60g) and the control group (80g). Each group was divided into four subgroups according to the time of exertion. The subgroups were stained with hematoxylin and eosin by serial sections. The root resorption was observed by tartrate-resistant acid phosphatase staining and the expression of interleukin-17 was observed by immunohistochemical method and double antibody sandwich enzyme-linked immunosorbent assay (ELISA). Results. 1) the root resorption region was mainly expressed in 1 / 3 of the root, and the root resorption occurred at the 4th day after the same force and reached the peak at the 7th day, and gradually decreased at the 14th day. Dentin repair appeared in some root resorption lacunae, and the degree of root resorption increased with the increase of orthodontic force. 2) No positive expression of interleukin-17 was found in the pseudo-experimental group. The expression of interleukin-17 was mainly weak positive on the 1st day, and increased with time, and reached the peak at the 7th day, and showed strong positive at the 7th day. Then to the 14th day, the expression intensity gradually decreased, and the expression was moderately positive. The greater the force value was at the same time, The stronger the positive expression was, the more significant the correlation between the expression of interleukin-17 and the time of exertion and the value of force was significant (P 0.05), and positive correlation was found between the expression of interleukin-17 and the value of force. 3) there was a positive correlation between the root resorption area and the positive rate of interleukin-17 expression under the conditions of different applied time and force value, which had statistical significance. Conclusion. 1) the degree of root resorption was regular at different applied time and force value. The greater the force value was, the more serious the root resorption was, and the root resorption degree increased from the 4th day to the 7th day, and reached the peak value at 7 days. At the 14th day, dentin repair appeared in the root resorption area. 2) the change of interleukin-17 is the same as the change of root absorption. Interleukin-17 is involved in the process of orthodontic periodontal tissue remodeling, which suggests that attention should be paid to the control of stress and cycle in clinical operation. 3) Interleukin-17 may be involved in the root resorption process of orthodontic teeth.
