脂肪干细胞的培养鉴定及多向诱导分化

【摘要】:目的:进行脂肪干细胞的分离、培养和鉴定,阐明脂肪干细胞的生物学特性。方法:无菌条件下取新西兰大耳白兔腹股沟处脂肪组织,从中获得纯化的脂肪来源干细胞。HE染色观察细胞形态;流式细胞术检测细胞表面标志物;多向诱导后检测其成脂、成骨及成软骨分化能力。结果:提取的兔脂肪干细胞形态为成纤维细胞样,生长旺盛,可以稳定增殖20代以上。流式细胞术分析显示CD44呈阳性表达,CD34、CD14呈阴性表达;经体外诱导培养后,向成脂细胞、成骨细胞及成软骨细胞分化。结论:本实验成功分离、培养和鉴定了脂肪干细胞,脂肪干细胞体外增殖稳定,并且具有成脂、成骨及成软骨多向分化潜能。
[Abstract]:Objective: to isolate, culture and identify adipose stem cells and elucidate the biological characteristics of adipose stem cells.Methods: adipose tissue in inguinal region of New Zealand white rabbit was harvested under aseptic condition and purified adipose-derived stem cells were obtained. He staining was used to observe cell morphology, flow cytometry was used to detect cell surface markers, and lipid production was detected after multi-directional induction.Ability of osteogenesis and cartilage differentiation.Results: the extracted adipose stem cells were fibroblast-like and could proliferate steadily for more than 20 generations.Flow cytometry showed that the expression of CD44 was positive and CD34 + CD14 was negative, and differentiated into adipoblasts, osteoblasts and chondroblasts after induction and culture in vitro.Conclusion: adipose stem cells were isolated, cultured and identified successfully in this experiment. Adipose stem cells proliferate stably in vitro and have the potential of adipogenic, osteogenic and cartilage differentiation.
