FPS-ZM1拮抗晚期糖基化终末产物受体逆转高糖所致的牙周膜成纤维细胞炎症反应

【摘要】:目的:以体外培养的人牙周膜成纤维细胞(hPDLFs)为研究对象,探讨新型小分子RAGE特异性抑制剂对高糖环境培养下牙周膜成纤维细胞炎性因子的影响。方法:原代培养hPDLFs,Western-Blot检测高糖对RAGE表达的影响,RT-PCR检测FPS-ZM1对高糖环境下IL-6和TNF-α基因表达的影响,ELISA检测IL-6和TNF-α的蛋白分泌变化。结果:高糖可导致hPDLFs表面RAGE表达增加,加重细胞炎症反应。而FPS-ZM1可以下调IL-6和TNF-α的基因表达,同时减少炎症介质IL-6和TNF-α的分泌。结论:RAGE特异性抑制剂FPS-ZM1可以在一定程度上逆转高糖刺激导致hPDLFs产生的炎症反应。
[Abstract]:Aim: to investigate the effect of a novel small molecule RAGE specific inhibitor on inflammatory cytokines of human periodontal ligament fibroblasts cultured in high glucose environment. Methods: the effect of high glucose on the expression of RAGE was detected by Western-Blot in primary culture. The effect of FPS-ZM1 on the expression of IL-6 and TNF- 伪 was detected by RT-PCR. The protein secretion of IL-6 and TNF- 伪 was detected by Elisa. Results: high glucose could increase the expression of RAGE on hPDLFs surface and aggravate the inflammatory reaction. FPS-ZM1 can down-regulate the gene expression of IL-6 and TNF- 伪, and decrease the secretion of IL-6 and TNF- 伪. Conclusion FPS-ZM1, a specific inhibitor of age, can reverse the inflammatory response of hPDLFs induced by high glucose stimulation to some extent.
