保留根面牙骨质对人牙周韧带细胞分化的影响

【摘要】:目的观察保留健康牙骨质对人牙周韧带细胞分化的影响。方法取正畸患者拔除的健康前磨牙,从牙根的近中和远中面釉牙骨质界下2 mm制备对称的5 mm×4 mm的牙片43对,随机分成观察组(保留牙骨质)和对照组(去除牙骨质)各43张。刮取牙周膜组织,按组织块贴壁法培养牙周韧带细胞,分离培养后,取两组各23张牙片,将细胞接种到牙片上共培养,制成PDLC-牙片复合体。7 d后刮除并收集PDLC-牙片复合体上的细胞,采用RT-PCR方法检测成牙骨质标记物牙骨质附着蛋白(CAP)和牙骨质蛋白23(CP-23)表达。取两组剩余的20张牙片制成细胞-牙片复合体放入半透明膜中,植入裸鼠体内,8周后比较两组牙片表面新形成的硬组织情况,并采用免疫组化方法观察骨桥蛋白(OPN)和骨唾液蛋白(BSP)表达。结果观察组CAP和CP-23表达较对照组升高(P均0.01)。植入裸鼠体内,8周后观察组14张牙片显示原有牙骨质上有牙骨质样基质形成;新旧牙骨质间并未观察到裂隙。对照组17张牙片在牙根表面有纤维组织形成,并且新纤维组织和牙本质表面间均可观察到不同宽度的裂隙。新形成的牙骨质样基质中,骨桥蛋白和骨唾液蛋白均呈阳性。结论保留健康牙根表面的牙骨质可能促进人牙周韧带细胞向成牙骨质细胞分化,有利于牙周组织愈合。
[Abstract]:Objective to observe the effect of preserving healthy cementum on the differentiation of human periodontal ligament cells. Methods healthy premolars extracted from orthodontic patients were randomly divided into two groups: the control group and the control group, and 43 pairs of symmetrical 5 mm 脳 4 mm dental slices were prepared from the proximal and distal enamel bone boundary 2 mm below the bone boundary of the tooth root, and were randomly divided into two groups: the observation group (reserved cementum) and the control group (removing cementum). Periodontal ligament cells were cultured according to the tissue block adherence method. After isolation and culture, 23 pieces of teeth were taken from each group, and the cells were co-cultured on the dental slices. The cells of PDLC- dentin complex were scraped and collected after 7. 7 d. The expression of cemental attachment protein (CAP) and cemental protein 23 (CP-23) were detected by RT-PCR. The cell-dental complex was made from the remaining 20 teeth of the two groups and put into the translucent membrane. After 8 weeks of implantation in nude mice, the newly formed hard tissues on the surface of the teeth were compared between the two groups. The expression of osteopontin (OPN) and bone saliva protein (BSPs) were observed by immunohistochemistry. Results the expression of CAP and CP-23 in the observation group was higher than that in the control group (P < 0.01). After implanted into nude mice for 8 weeks, 14 dental slices in the observation group showed cement-like matrix formation on the original cementum, but no fissure was observed between the new and old cementum. In the control group, there was fibrous tissue formation on the root surface of 17 teeth, and cracks of different widths could be observed between the new fibrous tissue and the dentin surface. Osteopontin and bone saliva were positive in newly formed cementoid matrix. Conclusion the preservation of cementum on the surface of healthy root may promote the differentiation of human periodontal ligament cells into cementoblasts, which is beneficial to the healing of periodontal tissue.
