Wnt1在口腔黏膜下纤维性变中的表达

【摘要】:目的:研究wnt1在口腔黏膜下纤维性变(OSF)患者治疗前后的表达并探讨其在OSF发生发展中的作用。方法:40例OSF中期患者采用曲安奈德+丹参局部封闭治疗4周,治疗前及第5周复诊时记录VAS及张口度,取唾液及龈沟液ELISA检测wnt1表达,实时荧光定量PCR检测OSF病变颊黏膜中wnt1 mRNA表达。结果:治疗前OSF组wnt1表达[颊黏膜定量PCR(36.89±10.40)×10~(-5),唾液浓度(61.61±4.45)ng/L,龈沟液浓度(56.20±3.65)ng/L]均高于正常组[颊黏膜定量PCR(4.63±1.53)×10~(-5),唾液浓度(40.26±3.00)ng/L,龈沟液浓度(53.45±1.74)ng/L](P0.01);OSF组经治疗后,唾液与龈沟液中wnt1表达均降低,且与OSF临床严重程度呈正相关(P0.05)。结论:wnt1可能参与OSF的发生发展,其在唾液与龈沟液中的实时监测有望成为评价OSF诊疗的无创性检控手段之一。
[Abstract]:Objective: to investigate the expression of wnt1 in patients with oral submucous fibrosis before and after treatment and its role in the development of OSF. Methods 40 middle stage OSF patients were treated with triamcinolone acetonide salvia miltiorrhiza for 4 weeks. VAS and mouth opening were recorded before and 5 weeks after treatment. Saliva and gingival crevicular fluid ELISA were taken to detect the expression of wnt1. The expression of wnt1 mRNA in buccal mucosa of OSF was detected by real-time fluorescence quantitative PCR. Results: before treatment, the expression of wnt1 [buccal mucosal quantitative PCR(36.89 卤10.40) 脳 10 ~ (-5) ng / L, saliva concentration (61.61 卤4.45 ng / L), gingival crevicular fluid concentration (56.20 卤3.65)ng/L)] in OSF group was higher than that in normal group [buccal mucosa quantitative PCR(4.63 卤1.53) 脳 10 ~ (-5) ~ (-5), saliva concentration was 40.26 卤3.00 ng / L, gingival crevicular fluid concentration was 53.45 卤1.74)ng/L] after treatment, the expression of wnt1 in saliva and gingival crevicular fluid decreased in OSF group. There was a positive correlation with the clinical severity of OSF (P 0.05). ConclusionWnt1 may be involved in the development of OSF and its real-time monitoring in saliva and gingival crevicular fluid is expected to be one of the non-invasive methods to evaluate the diagnosis and treatment of OSF.
