【摘要】:目的研究第10号染色体缺失的磷酸酶及张力蛋白同源基因蛋白(phosphatase and tensin homolog deleted on chromosome ten,PTEN)在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达及其临床意义。方法收集30例OSCC及30例距癌组织边界2 cm的癌旁组织,Western blot测定上述组织PTEN蛋白含量,qRT-PCR测定mRNA的表达水平,免疫组化(immunohistochemistry,IHC)定性测定PTEN蛋白在上述组织细胞中的组织学定位与表达情况,并根据PTEN表达结果分析其与OSCC临床病理关系。结果 1癌旁组织均为PTEN蛋白阳性表达,PTEN蛋白阳性表达主要为细胞浆内棕黄色或棕褐色染色。OSCC组织多为阴性表达。2OSCC癌组织中PTEN(蛋白和mRNA)的表达明显低于2 cm外的癌旁组织(P0.05)。3不同年龄、不同性别、不同发生部位OSCC的PTEN(蛋白和mRNA)表达差异无统计学意义(P0.05)。4PTEN在高分化鳞癌中的表达明显较中低分化鳞癌高(P0.05)。5无淋巴转移的较有淋巴转移的表达高(P0.05)。6临床Ⅰ+Ⅱ期表达明显高于Ⅲ+Ⅳ期(P0.05)。结论 PTEN在OSCC癌组织中存在不同程度的异常表达,可能是口腔鳞癌发生、发展、侵袭转移过程中的一个重要因素。
[Abstract]:Objective to study the expression and clinical significance of phosphatase and tensin homolog deleted on chromosome tenon (PTEN), a homologous protein of phosphatase and tension protein homologous gene, deleted on chromosome 10 in oral squamous cell carcinoma (OSCC). Methods 30 cases of OSCC and 30 cases of paracancerous tissues 2 cm from the margin of cancer were collected to detect the PTEN protein content and the expression of mRNA by qRT-PCR. Immunohistochemical staining (IHC) was used to determine the histological localization and expression of PTEN protein in these tissues. The clinicopathological relationship between IHC and OSCC was analyzed according to the results of PTEN expression. Results (1) the positive expression of PTEN protein was mainly brown or brown staining in the cytoplasm. The expression of PTEN (protein and mRNAs) was significantly lower than that outside 2 cm. P0. 05. 3 in adjacent tissues of cancer. Different sexes, There was no significant difference in the expression of PTEN (protein and mRNAs) between different sites of OSCC. The expression of P0.05N in highly differentiated squamous cell carcinoma was significantly higher than that in poorly differentiated squamous cell carcinoma (MSCC) without lymphatic metastasis.
