三维钛网支架与犬牙周膜成纤维细胞构建类牙周结构的实验研究

【摘要】:根据牙周膜成纤维细胞能向成牙骨质、成骨方向分化的多向分化潜能,,通过将犬牙周膜成纤维细胞混合细胞外基质,接种至三维钛网支架材料内部,植入裸鼠皮下,观察其形成类牙周结构的可能性。 利用胶原酶、胰蛋白酶消化组织块法原代培养获取犬牙周膜成纤维细胞,利用倒置显微镜、HE染色观察其细胞外形及生长方式,透射电镜观察细胞超微结构,通过免疫细胞化学染色对犬牙周膜进行来源鉴定,通过矿化结节实验证明其能向硬组织分化;用ECM gel混合犬牙周膜成纤维细胞到三维钛网支架材料内部,扫描电镜发现种子细胞在支架材料表面附着增殖程度较传统反复浸透法优良;硬组织切片发现有硬组织染色,且部分样本产生类似胶原纤维。
[Abstract]:According to the multidirectional differentiation potential of periodontal ligament fibroblasts into cementoblasts and osteoblasts, the canine perodontic ligament fibroblasts were mixed with extracellular matrix and inoculated into the three-dimensional titanium mesh scaffold and implanted subcutaneously into nude mice. The possibility of periodontal formation was observed. The canine periodontic membrane fibroblasts were obtained by using collagenase and trypsin digestion tissue block method. The morphology and growth pattern of the cells were observed by HE staining with inverted microscope, and the ultrastructure of the cells was observed by transmission electron microscope (TEM). The origin of canine periodontic membrane was identified by immunocytochemical staining, and it was proved by mineralized nodules that it could differentiate into hard tissue. Scanning electron microscopy showed that seed cells had better adhesion and proliferation on the surface of scaffolds than the traditional repeated immersion method. Hard tissue staining was found in hard tissue sections and some samples produced collagen-like fibers.
