血管内皮细胞生长因子与碱性成纤维细胞生长因子联合应用对鼠牙周膜成纤维细胞增殖与碱性磷酸酶活性的影响
[Abstract]:Background: basic fibroblast growth factor can improve fibroblast proliferation and collagen deposition. Vascular endothelial growth factor can improve the blood flow perfusion and metabolism of pathological tissue at appropriate concentration. The activity of alkaline phosphatase could be enhanced under the action of time. Aim: to observe the effects of vascular endothelial growth factor (VEGF) combined with basic fibroblast growth factor (bFGF) on proliferation and alkaline phosphatase activity of rat periodontal ligament fibroblasts. Methods: (1) Rat periodontal ligament fibroblasts were cultured in vitro and their embryonic origin was identified. Effects of basic fibroblast growth factor on proliferation of periodontal ligament fibroblasts in rats and determination of the apparent and maximum effective concentrations. (3) the cells were divided into five groups: group A: control group. Group B: Vascular endothelial growth factor maximum effect concentration group C: basic fibroblast growth factor maximum effect concentration group D group: vascular endothelial growth factor concentration Group E: the maximum effect concentration of vascular endothelial growth factor and the maximum effect concentration of basic fibroblast growth factor. The alkaline phosphatase activity of 5 groups of cells was measured after 14 days. Results and conclusion: (1) the cultured rat periodontal ligament fibroblasts grew well and originated from mesoderm. (2) with the increase of vascular endothelial growth factor, the concentration of basic fibroblast growth factor increased. The proliferation ability of rat periodontal ligament fibroblasts was also increased (P0.01). The apparent concentration of vascular endothelial growth factor is 10 渭 g / L, the maximum effective concentration is 100 渭 g / L; the apparent concentration of basic fibroblast growth factor is 0.1 渭 g / L, and the maximum effective concentration is 10 渭 g / L; (3) when vascular endothelial growth factor, When combined with basic fibroblast growth factor (bFGF) on periodontal ligament fibroblasts of rats, the A value of each group was higher than that of the control group. (4) after 3 and 7 days of treatment, the A value of each group was higher than that of the control group. However, the value of A in group D was significantly lower than that in group E (P0.05) for 14 days. (5) the results showed that the combined use of maximum effect concentration could play an obvious synergistic effect on the activity of alkaline phosphatase in a certain time, but beyond a certain time. There was no difference between the apparent concentration and the maximum effect concentration on alkaline phosphatase activity of periodontal ligament fibroblasts. This may be related to the aging of two factors and the receptor of periodontal ligament cells.
