不同转移潜能SD大鼠舌鳞状细胞癌细胞株的分离及生物学特性比较

【摘要】：目的：获得高转移舌鳞状细胞癌细胞,为建立高转移舌癌动物模型奠定基础。方法：采用改良侵袭实验筛选出高低不同的转移潜能细胞；采用MTT实验和平板克隆实验检测两种细胞的克隆差异；侵袭性实验和迁移性实验检测两种细胞侵袭和转移差异；流式细胞仪用来检测两种细胞生长周期的差异；采用Western Blot和Real-time PCR方法检测两种细胞Zeste基因增强子同源物2(EZH2)的蛋白和mRNA表达差异。结果：分离出两种不同转移潜能细胞。两种不同转移细胞的克隆形成、侵袭和迁移、细胞周期和EZH2蛋白和mRNA表达均有显著性差异(t=3.423～59.300,P0.05～0.001)。结论：从舌鳞癌细胞系中分离得到了两种不同转移潜能的细胞亚群,且两种细胞具有不同的生物学特性。
[Abstract]:Objective: to obtain high metastatic squamous cell carcinoma of tongue and to establish animal model of high metastasis of tongue cancer. Methods: different metastatic potential cells were screened by modified invasion assay, clone differences were detected by MTT assay and plate cloning assay, invasion and metastasis were detected by invasive assay and migration assay. Flow cytometry was used to detect the difference of cell growth cycle, and Western Blot and Real-time PCR methods were used to detect the protein and mRNA expression of Zeste gene enhancer homologue 2 (EZH2). Results: two different metastatic potential cells were isolated. There were significant differences in cell cycle, EZH2 protein and mRNA expression between the two kinds of metastatic cells. Conclusion: two kinds of cell subsets with different metastatic potential were isolated from tongue squamous cell carcinoma cell lines, and the two kinds of cells had different biological characteristics.
