Foxo3a调控小鼠釉质发育过程中MMP-20基因的表达

【摘要】:目的:探讨转录因子Foxo3a在釉质发育过程中调控基质金属蛋白酶-20(MMP-20)基因表达的分子机制。方法:分别利用免疫组织化学技术、免疫荧光、RT-PCR法、双荧光素酶报告基因检测系统检测转录因子Foxo3a与MMP-20启动子区域的转录调控水平,分析Foxo3a调控MMP-20的转录活性的机制。结果:免疫组织化学染色发现Foxo3a在成釉细胞核中呈阳性表达,并且随着釉质的分泌表达逐渐增强,晚期变弱;RT-PCR证实Foxo3a促进MMP-20基因表达(P0.05);双荧光素酶检测显示,Foxo3a促进MMP-20表达(P0.05);特定序列定点突变后的Foxo3a对MMP-20基因的转录活性无显著的影响(P0.05)。结论:转录因子Foxo3a可能通过与特定序列结合而调控MMP-20的表达,从而在釉质分泌过程中发挥重要作用。
[Abstract]:Aim: to investigate the molecular mechanism of transcription factor Foxo3a regulating matrix metalloproteinase-20 (MMP-20) gene expression during enamel development. Methods: immunohistochemistry, immunofluorescence RT-PCR and double luciferase reporter gene detection system were used to detect the transcriptional regulation level of Foxo3a and MMP-20 promoter regions, and to analyze the mechanism of Foxo3a regulating the transcriptional activity of MMP-20. Results: immunohistochemical staining showed that Foxo3a was positively expressed in the ameloblast nucleus and increased with the secretory expression of enamel. RT-PCR confirmed that Foxo3a promoted the expression of MMP-20 gene (P0.05); double luciferase assay showed that Foxo3a promoted MMP-20 expression (P0.05); Foxo3a with specific sequence site-directed mutation had no significant effect on the transcriptional activity of MMP-20 gene (P0.05). Conclusion: transcription factor Foxo3a may play an important role in the process of enamel secretion by regulating the expression of MMP-20 by binding to specific sequences.
