单核细胞和巨噬细胞中IL-18与颞下颌关节炎的相关性研究

【摘要】:目的通过检测单核细胞和巨噬细胞上IL-18R的表达,探讨IL-18在鼠颞下颌关节炎中的可能作用。方法SPF级BALB/c雄性小鼠30只,随机分为A、B、C、D、E组,每组六只。每组给予不同处理:A组仅注射0.9%氯化钠注射液、B组注射Ⅱ型胶原酶和NS;C组注射Ⅱ型胶原酶和IL-18;D组注射Ⅱ型胶原酶和IL-18BP;E组注射Ⅱ型胶原酶和IL-18BP和IL-18。B、C、D、E组分别在第1天和第21天时注射Ⅱ型胶原酶(2 g/ml)100μl;C、D、E组每次注射100μl IL-18/IL-18BP(10 ng/ml),每三天注射一次。第25天,采集各组小鼠外周血、组织灌洗液。采用流式细胞术检测单核细胞和巨噬细胞中IL-18受体阳性表达变化,酶联免疫吸附实验法检测血浆和关节组织灌洗液上清液中炎症因子TNF-α、IL-1β水平。初步探究在单核细胞和巨噬细胞中IL-18在颞下颌关节炎发病机制中起到的作用。结果1.流式细胞术中B、C、D、E组小鼠外周血中单核细胞和颞下颌灌洗液中巨噬细胞上IL-18R的阳性表达显著高于A组(p㩳0.001);单核细胞和巨噬细胞上IL-18R的表达C组明显高于B、D、E组(p㩳0.001)。2.酶联免疫吸附实验中B、C、D、E组小鼠血浆和颞下颌关节灌洗液上清中炎症因子TNF-α和IL-1β的水平均高于A组(p㩳0.001);C组中炎症因子TNF-α和IL-1β的水平明显高于B、D、E组(p㩳0.001)。结论鼠颞下颌关节炎模型中单核细胞和巨噬细胞上IL-18R的表达升高,IL-18可诱导IL-18R表达进一步升高,提示IL-18可能参与颞下颌关节炎发病。IL-18BP可抑制IL-18诱导的IL-18R的表达和炎症因子的释放,提示IL-18可能是治疗颞下颌关节炎的潜在靶点。
