下调α-catulin基因的表达对舌鳞癌细胞株Tscca侵袭及迁移能力影响的体外研究

【摘要】:目的探讨α-catulin基因下调对人舌鳞癌细胞株Tscca侵袭、迁移能力的影响。方法本实验设为3个组:α-catulin-siRNA组、空质粒转染组和未转染质粒的空白对照组。构建α-catulin沉默质粒转染的人舌鳞癌细胞株Tscca,运用Western blotting检测α-catulin蛋白表达,采用Transwell及划痕实验检测细胞的侵袭、迁移能力。结果 Western blotting结果显示,α-catulin-siRNA转染组Tscca细胞α-catulin蛋白表达量减少;Transwell及划痕实验结果显示,α-catulin-siRNA转染组细胞侵袭、迁移能力降低。结论下调α-catulin基因的表达可以有效降低人舌鳞癌细胞株Tscca的侵袭、迁移能力,α-catulin有望成为舌癌基因治疗的一个潜在靶点。
[Abstract]:Objective to investigate the effect of down-regulation of 伪-catulin gene on the invasion and migration of human tongue squamous cell carcinoma cell line Tscca. Methods the experiment was divided into three groups: 伪-catulin-siRNA group, empty plasmid transfection group and blank control group. Human tongue squamous cell carcinoma cell line Tscca, transfected with 伪-catulin silencing plasmid was constructed to detect the expression of 伪-catulin protein by Western blotting, and the invasion and migration ability of the cells were detected by Transwell and scratch test. Results the results of Western blotting showed that the expression of 伪-catulin protein decreased in Tscca cells transfected with 伪-catulin-siRNA, and the expression of 伪-catulin protein in Tscca cells was decreased by Transwell and scratch test. The results of Transwell and scratch test showed that the cell invasion and migration ability of 伪-catulin-siRNA transfected group were decreased. Conclusion down-regulation of 伪-catulin gene expression can effectively reduce the invasion and migration ability of human tongue squamous cell carcinoma cell line Tscca. 伪-catulin may be a potential target of tongue oncogene therapy.
