OLP患者外周血T细胞B7-H1及PD-1的表达情况及与疾病状态的关系研究

【摘要】：目的:通过检测口腔扁平苔藓(oral lichen planus,OLP)患者外周血T细胞B7-H1及PD-1的表达情况及与疾病状态的关系,以此来探讨B7-H1/PD-1信号通道在OLP患者T细胞免疫中的作用。方法:选择22例OLP患者,12例健康成人作为对照。应用REU计分系统及视觉模拟评分法(VAS)对OLP患者的疾病状态进行计分。密度梯度离心法分离OLP组和对照组外周血中的单个核细胞,流式细胞术检测单个核细胞中T细胞表面B7-H1及PD-1的表达水平。贴壁法去除单核细胞后,将OLP患者的淋巴细胞与健康成人淋巴细胞混合培养,加入抗B7-H1单克隆抗体阻断B7-H1/PD-1信号通道,培养5d后应用XTT方法检测细胞增殖情况。结果:CD3+B7-H1+、CD3+PD-1+的表达百分率在OLP和正常对照组之间存在有明显差异(P0.05)。CD3+B7-H1+在OLP组的表达水平与疾病状态计分正相关(P=0.002),而CD3+PD-1+的表达水平与疾病的临床状态计分无关(P=0.537)。在混合淋巴细胞培养中,使用抗B7-H1单克隆抗体阻断B7-H1/PD-1信号通道后,淋巴细胞增殖情况与对照组无明显差异(P0.05)。结论:OLP患者外周血T细胞中的CD3+B7-H1+、CD3+PD-1+很可能参与了OLP的发病;CD3+B7-H1+对OLP的疾病进展可能产生一定作用。
[Abstract]:Objective: to detect the expression of B7-H1 and PD-1 in peripheral blood T cells of patients with oral lichen planus (oral lichen planus,OLP) and its relationship with disease status. To explore the role of B7-H1/PD-1 signal channel in T cell immunity of OLP patients. Methods: 22 patients with OLP and 12 healthy adults were selected as control group. REU scoring system and visual analogue score (VAS) were used to score the disease status of OLP patients. Mononuclear cells were isolated from peripheral blood of OLP group and control group by density gradient centrifugation. The expression levels of B7-H1 and PD-1 on T cell surface were detected by flow cytometry. After the monocytes were removed by adherent method, lymphocytes from OLP patients were mixed with healthy adult lymphocytes, and monoclonal antibodies against B7-H1 were added to block the signal channels of B7-H1/PD-1. After 5 days of culture, the proliferation of cells was detected by XTT method. Results: there was significant difference in the expression of CD3 B7-H1 and CD3 PD-1 between OLP and normal control group (P0.05). The expression level of CD3 B7-H1 in OLP group was positively correlated with the score of disease state (P0. 002). However, the expression of CD3 PD-1 was not correlated with the clinical status score (P0. 537). In mixed lymphocyte culture, there was no significant difference in lymphocyte proliferation between the control group and the control group after blocking the B7-H1/PD-1 signal channel with anti B7-H1 monoclonal antibody (P0.05). Conclusion: CD3 B7-H1 and CD3 PD-1 in peripheral blood T cells of OLP patients may be involved in the pathogenesis of OLP, and CD3 B7-H1 may play a role in the progression of OLP.
