顺铂调控舌鳞癌细胞增殖和迁移能力的研究

【摘要】:目的:探讨舌鳞癌细胞(human tongue squamous cell carcinoma cells,SCC)经顺铂化疗后的增殖和迁移能力的改变,及其对细胞增殖相关Nanog基因表达的调控作用。方法:本文应用5、4、3.5μg/m L顺铂分别作用于舌鳞癌细胞系SCC-15,处理时间分别为24、48、72 h(实验组),未经顺铂处理的细胞为对照。流式细胞仪检测细胞凋亡率,q RT-PCR及Western blot检测2组细胞中Nanog的表达情况,划痕实验检测2组细胞迁移能力的变化。结果:经顺铂处理后细胞凋亡率增加,48 h后凋亡率显著增加(P=0.000)。q RT-PCR结果显示24、48、72 h组的Nanog基因相对表达量在顺铂处理的细胞中逐渐增加,48 h Nanog基因表达量与对照组相比,差异具有统计学意义(P=0.000)。Western blot实验结果进一步验证了顺铂处理的舌鳞癌细胞中Nanog蛋白相对表达量较对照组高(P≤0.001)。划痕实验显示经顺铂作用时间越长的实验组,细胞迁移速率越高。结论:顺铂化疗对舌鳞癌细胞增殖能力和迁移能力有调控作用。
[Abstract]:Aim: to investigate the effects of cisplatin chemotherapy on proliferation and migration of tongue squamous cell carcinoma (human tongue squamous cell carcinoma cells,SCC) cells and its regulatory effect on the expression of Nanog gene.
