紫杉醇对裸鼠体内涎腺腺样囊性癌的抑制作用

【摘要】：目的:观察紫杉醇对裸鼠体内涎腺腺样囊性癌移植瘤的抑制作用。方法:按常规将涎样囊性癌ACC-M细胞接种于裸鼠皮下,2周后将荷瘤鼠随机分为2组(n=8),对治疗组鼠皮下注射紫杉醇,10μg/g,1次/d。对照组鼠给予等量生理盐水。定期测量肿瘤体积,治疗2周后处死,测量肿瘤重量,病理学观察,流式细胞术检测各组凋亡率。免疫组化法检测血管内皮生长因子(VEGF)在肿瘤细胞中的表达。结果:治疗2周后紫杉醇治疗组抑瘤率42.74%,治疗组和对照组肿瘤细胞凋亡率分别为(24.3±2.1)%和(9.2±1.9)%;VEGF蛋白表达平均吸光度值(MOD)分别为0.322±0.033和0.388±0.029(P0.01)。结论:紫杉醇能够抑制ACC-M细胞在裸鼠体内的增殖,促进其凋亡,抑制VEGF的表达。
[Abstract]:Aim: to observe the inhibitory effect of paclitaxel on adenoid cystic carcinoma of salivary gland in nude mice. Methods: ACC-M cells of salivary cystic carcinoma were subcutaneously inoculated into nude mice. After 2 weeks, tumor bearing mice were randomly divided into two groups (n = 8). Paclitaxel was injected subcutaneously into the treatment group with 10 渭 g / g paclitaxel once a day. Rats in the control group were given the same amount of normal saline. Tumor volume was measured regularly and killed after 2 weeks of treatment. Tumor weight, pathological observation and apoptosis rate were measured by flow cytometry. The expression of vascular endothelial growth factor (VEGF) in tumor cells was detected by immunohistochemistry. Results: after 2 weeks of treatment, the tumor inhibition rate of paclitaxel group was 42.74%, and the apoptosis rate of tumor cells in treatment group and control group was (24.3 卤2.1)% and (9.2 卤1.9)%, respectively. The average absorbance of VEGF protein expression was 0.322 卤0.033 and 0.388 卤0.029, respectively (P0.01). Conclusion: paclitaxel can inhibit the proliferation of ACC-M cells in nude mice, promote their apoptosis and inhibit the expression of VEGF.
