上海出入境检验检疫局发现我国首例库波热病毒

【摘要】:针对西非安哥拉回国的不明原因发热病人,筛查其可能携带的传染病病原体。采用荧光PCR的方法对口岸重点关注的黄热病毒、寨卡病毒、登革病毒等病原体进行筛查;采用深度测序的方法,搜索病毒库、细菌库、寄生虫库,进行序列比对筛查各类病原体,采用全基因组测序的方法获得病原体全基因组序列。2017年1月2日凌晨1点,上海出入境检验检疫局国家国境口岸卫生监督检测重点实验室接到一份紧急样本,样本来自1名安哥拉回国的28岁男子。该男子在安哥拉当地医院初筛黄热病阳性,该男子要求自行回国治疗。针对病人的血液样本和尿液样本,开展黄热病毒、登革病毒、寨卡病毒、裂谷热病毒、基孔肯雅病毒、西尼罗病毒、流行性乙型脑炎病毒、疟原虫等多种虫媒病原体的荧光PCR检测,结果显示阴性。通过对血液样本的深度测序发现,该病人感染了一种新型的弹状病毒,中文命名为库波热病毒,并获得了库波热病毒全基因组序列,与尼日利亚发现的库波热病毒比较,同源性为96%。
[Abstract]:Screening of infectious disease pathogens for unknown fever patients returning from Angola in West Africa.Fluorescent PCR was used to screen yellow fever virus, Zika virus, dengue virus and so on, and deep sequencing method was used to search virus library, bacterial bank, parasite bank, etc.Sequence alignment was carried out to screen all kinds of pathogens, and the whole genome sequence was obtained by whole genome sequencing. 1: 00 am on January 2, 2017,Shanghai Entry-Exit Inspection and Quarantine Bureau has received an emergency sample from a 28-year-old man who returned home from Angola.The man was tested positive for yellow fever at a local hospital in Angola and asked to return home for treatment.Yellow fever virus, dengue virus, Zika virus, Rift Valley virus, Kikungunya virus, West Nile virus, Japanese encephalitis virus,Fluorescent PCR analysis of malaria parasite and other arbogenic pathogens showed negative results.By deep sequencing of blood samples, it was found that the patient was infected with a new type of slug virus, named Kubovirus in Chinese, and obtained the whole genome sequence of Kubovirus, which was compared with the Kubovirus found in Nigeria.The homology is 96.
