白芍木瓜汤防治大鼠骨性关节炎的实验研究

【摘要】:目的:建立骨性关节炎大鼠模型,用白芍木瓜汤治疗干预,观察白芍木瓜汤对OA大鼠一般情况、体重、抓力、影像学和病理学表现的影响,验证白芍木瓜汤对OA大鼠防治作用,并探讨其对骨性关节炎的可能作用机制。方法:清洁级雄性SD大鼠,共50只,将其随机分成5组:正常对照组(A组)、模型对照组(B组)、阳性对照组(C组)、白芍木瓜汤高剂量组(D组)、白芍木瓜汤低剂量组(E组),每组10只。其中正常对照组不予任何处理,其余4组大鼠均采用双侧膝关节腔内注射碘乙酸法,成功建立OA大鼠模型。术后第二天,A、B组给予生理盐水灌胃,C组给予布洛芬灌胃,D、E两组采用不同剂量的中药汤剂灌胃,持续灌胃6周,在此期间,每周称量各组大鼠的体重及测定其抓力程度。至第6周末,对各组大鼠行腹腔麻醉,用X-ray影像观察膝关节结构与状态;心脏采血,并分离血清,用酶联免疫(Elisa)法测定各组大鼠的血清中基质金属蛋白酶13(MMP-13)、软骨蛋白聚糖(Aggrecan)、Ⅱ型胶原(Col Ⅱ)含量;并截取膝关节,剔除皮肤与肌肉,EDTA脱钙后分别进行HE染色与SO染色,光镜下观察各组大鼠膝关节病理变化。结果:(1)体重:造模成功后,B组的大鼠体重相对于A组,始终存在显著性差异(P0.01),随着给药时间的增加,白芍木瓜汤可以改善大鼠食欲,使大鼠体重增加。(2)抓力测定:各治疗组的大鼠抓力水平始终无明显差异(P0.05)。(3)膝关节影像:造模成功后,相对于A组,B组大鼠的双膝关节破坏严重,干预治疗6周后,C、D、E组大鼠双膝关节的影像学表现均明显改善,以D组的改善情况最为明显。(4)膝关节病理:造模成功后,相比于A组,B组的大鼠双膝关节破坏程度高;干预治疗6周后,相比于B组,C、D、E组膝关节软骨的病理学均明显改善,以D组的改善情况最为明显。(5)血清含量:造模成功后,相对于A组,B组大鼠血清中的MMP-13含量呈显著性升高(P0.01),Col-Ⅱ、Aggrecan含量呈显著性降低(P0.01);药物治疗6周后,相比于B组,C、D、E组大鼠血清中的MMP-13含量均呈显著性降低(P0.01),血清Col-Ⅱ、Aggrecan含量均呈显著性升高(P0.01)。结论:白芍木瓜汤对碘乙酸诱导的大鼠骨性关节炎有良好的药效作用,其作用机制可能与抑制MMP-13产生,促进Aggrecan、Col Ⅱ合成,从而达到修复软骨有关。
[Abstract]:Objective: to establish a rat model of osteoarthritis, and to observe the effects of paeonia-papaya decoction on the general condition, body weight, grasping ability, imaging and pathology of OA rats, and to verify the preventive and therapeutic effects of paeonia-papaya decoction on OA rats. To explore the possible mechanism of its action on osteoarthritis. Methods: fifty clean grade male SD rats were used. They were randomly divided into 5 groups: normal control group (group A), model control group (group B), positive control group (group C), high dose group of paeoniae papaya decoction (group D) and low dose group of paeoniao papaya decoction (group E) with 10 rats in each group. The normal control group was not treated with any treatment, and the other four groups were successfully established OA rat model by injecting iodoacetic acid into bilateral knee joint. On the second day after operation, normal saline was given in group A and ibuprofen was given in group C. The rats in group D were given different doses of traditional Chinese medicine decoction for 6 weeks. During this period, the weight of rats in each group was measured weekly and the degree of grasping was measured. At the end of the 6th week, the rats in each group were anesthetized intraperitoneally, the structure and state of knee joint were observed by X-ray imaging, the blood was taken from the heart, and the serum was separated.
