酶-二氧化锰纳米片杂化探针自指示比色传感葡萄糖

【摘要】:构建一种二氧化锰纳米片与葡萄糖氧化酶杂化自指示比色探针,在弱酸性条件下,葡萄糖氧化酶专属性催化氧化葡萄糖,定量产生葡萄糖酸和过氧化氢,生成的过氧化氢有效分解二氧化锰纳米片,探针体系特征吸收波长(374 nm)处吸收度降低。吸收度变化量与葡萄糖浓度呈线性相关,线性范围1~20μmol/L,相关系数R~2=0.990 1,检测限低至0.1μmol/L。探针对葡萄糖具有高选择性和灵敏度,血清样品中常见的氨基酸、阴离子、阳离子及蛋白质等小分子物质造成干扰均可忽略,实际样品测定具有前处理简单、操作方便、快速等特点。此外,鉴于二氧化锰的氧化还原特性以及酶催化体系的多样性,酶-二氧化锰纳米片杂化探针平台具有普适性,可设计构建多种分析物的比色传感器。
[Abstract]:A hybrid self-directed colorimetric probe of manganese dioxide nanoparticles and glucose oxidase was constructed. Glucose oxidase was used to catalyze the oxidation of glucose to quantitatively produce gluconic acid and hydrogen peroxide under the condition of weak acidity. The resulting hydrogen peroxide effectively decomposes manganese dioxide nanoparticles and decreases the absorption at the characteristic absorption wavelength of the probe system at 374 nm. There was a linear correlation between the absorbency and glucose concentration. The linear range was 1 ~ 20 渭 mol / L, the correlation coefficient was RV 20.990 1, and the detection limit was as low as 0.1 渭 mol 路L ~ (-1). The probe has high selectivity and sensitivity to glucose. The interference caused by small molecules such as amino acids, anions, cations and proteins in serum samples can be ignored. Speed, etc. In addition, in view of the redox characteristics of manganese dioxide and the diversity of enzyme catalytic system, the hybrid probe platform of enzyme manganese dioxide nanochip is universal and can be used to design and construct colorimetric sensors for many kinds of analytes.
