姜黄素对干热环境下大鼠肾损伤保护作用的研究

【摘要】:目的:通过姜黄素预处理大鼠并复制干热环境下大鼠中暑模型,探讨姜黄素对干热环境下大鼠肾损伤的保护作用及其可能机制。方法:成年SD雄性大鼠200只,随机数字表法分为四:0分钟组(常温组)、50分钟组、100分钟组、150分钟组。每组下分为五个亚组(n=10):盐水(NS)组、溶剂(CMCNa)组、姜黄素50mg/kg组(Cur 50mg/kg)、姜黄素100mg/kg组(Cur 100mg/kg)、姜黄素200mg/kg组(Cur 200mg/kg)。盐水组给予0.9%生理盐水灌胃,溶剂组给予0.5%CMCNa溶液灌胃,姜黄素组大鼠给予不同浓度姜黄素溶液灌胃,连续七天,每日一次。复制干热环境下大鼠中暑模型:将大鼠置于模拟沙漠干热环境中(西北特殊环境人工实验舱,中国人民解放军乌鲁木齐总医院研制),设置温度41±0.5℃,湿度10±1%,每25分钟监测大鼠核心体温。根据分组时间取出大鼠,进行麻醉,腹主动脉采血进行生化检测,穿刺膀胱留取尿液进行肾损伤分子-1(KIM-1)及中性粒细胞明胶酶相关载脂蛋白(NGAL)的检测,取肾组织用于病理及分子生物学检测。结果:(1)在50min、100min及150min组,不同浓度的姜黄素处理后的大鼠血清中尿素、肌酐、KIM-1及NGAL的含量较同一时间点的盐水(NS)组和溶剂组(CMCNa)明显下降(P0.05)。(2)在干热环境中,不同浓度的姜黄素可降低肾组织中MDA含量及提高SOD和CAT的活性,差异有统计学意义(P0.05)。(3)在同一时间点干热环境下,姜黄素预处理组大鼠肾组织内IL-1β、IL-6和TNF-α的水平较盐水(NS)组及溶剂(CMCNa)组降低,差异具有统计学意义(P0.05)。(4)在150min组,不同浓度的姜黄素预处理可明显改善大鼠肾组织的病理改变。(5)大鼠肾组织石蜡切片TUNEL染色显示,在150min组,姜黄素预处理可降低肾脏细胞的凋亡率,差异具有统计学意义(P0.05)。(6)在150min组,姜黄素预处理可降低肾组织内炎症反应相关蛋白及凋亡相关蛋白的表达水平,差异具有统计学意义(P0.05)。结论:(1)持续的干热环境对大鼠肾脏具有明显的损伤作用,并且随着持续时间的延长而加重,肾组织内炎症反应及氧化应激水平升高,肾组织细胞凋亡显著。(2)干热环境下,不同浓度的姜黄素均可不同程度的减轻大鼠肾损伤,其中200mg/kg时效果最显著。(3)姜黄素可能通过抑制炎症及氧化应激反应、阻断细胞凋亡通路,在一定程度上减轻了干热环境下大鼠肾损伤。
[Abstract]:Aim: to investigate the protective effect of curcumin on renal injury and its possible mechanism by pretreating rats with curcumin and making heat stroke model in dry heat environment. Methods: 200 adult Sprague-Dawley male rats were randomly divided into 4: 0 minute group (normal temperature group), 50 minutes group, 100 minutes group and 150 minutes group. Each group was divided into five subgroups: saline group (NS), solvent group (CMCNa), curcumin 50mg/kg group (curcumin 50mg/kg), curcumin 100mg/kg group (curcumin 100mg/kg), curcumin 200mg/kg group (curcumin 200mg/kg). The saline group was given 0.9% normal saline, the solvent group was given 0.5 CMCNa solution, and the curcumin group was given curcumin solution of different concentrations once a day for seven days.
