1型糖尿病视网膜病变大鼠脂联素水平与TGF-β1及ICAM-1的关系

【摘要】:目的:糖尿病视网膜病变(DR)是其并发症中最为常见和严重的,是成人致盲的疾病之一。世界范围内糖尿病患者人数的不断增大,使DR成为了视力丧失以及视功能损害的主要原因。研究脂联素(ADPN)与l型糖尿病大鼠模型的视网膜组织中转化生长因子-β1及细胞间粘附分子-1之间的关系,以及各个因子分别与DR的关系。方法:选取清洁级、近交SD(Sprague Dawley)雄性大鼠40只(体重介于180g-220g)分为2组,正常组(NC组,n=16),糖尿病组(DM组,n=24)。DM组的大鼠给予腹腔注射STZ(按计量为65mg/kg)一次足量给药,注射药物STZ 72h后测DM组大鼠血清Glu≥16.7mmol/L,出现尿频、饮水及食量增多的症状,即大鼠T1DM模型制作成功。NC组大鼠用等计量的枸橼酸钾的缓冲溶液,腹腔一次性给药。2组大鼠均给予一般饮食,于相同的情况下观察7日。模型组始终满足1型糖尿病(T1DM)模型的条件,未见转复正常的纳入实验组。后2组大鼠均继续给予普通饲料喂养,分别观察4周及8周时2组大鼠视网膜病变的程度、组织中TGF-β1、ICAM-l及血清中ADPN的浓度。根据实验结果以及监测数据的分析和比较,讨论它们的关系。结果:l、T1DM大鼠的血清中ADPN含量的水平明显较正常时降低,且随着病程时间的延长ADPN的水平越低。2、视网膜组织中的转化生长因子-β1及细胞间粘附分子-1的表达随着糖尿病病程延长,组织间表达明显增加。3、视网膜组织病变的严重程度是伴随T1DM病程的延长,逐渐加重的。结论:1型糖尿病大鼠的血清脂联素水平是随着病程的发展而逐渐减低的,并且与正常组比较其水平是明显下降的。视网膜组织中TGF-β1及ICAM-1的表达随病程的进展是明显升高的,纤维化程度明显加重。可认为随着脂联素的减少TGF-β1及细胞间粘附分子-1的表达逐渐增加,视网膜病变程度更高。
[Abstract]:Objective: diabetic retinopathy (DR) is one of the most common and severe complications of diabetic retinopathy. With the increasing number of diabetic patients worldwide, DR has become the main cause of visual loss and visual impairment. To study the relationship between adiponectin (ADPN) and transforming growth factor- 尾 1 (TGF- 尾 1) and intercellular adhesion molecule-1 (ICAM-1) in retina of type 1 diabetic rats, and the relationship between each factor and DR. Methods: forty inbred SD (Sprague Dawley) male rats (weight between 180g-220g and clean grade) were divided into two groups: normal group (NC group, nong16), diabetic group (DM group), and normal control group (NC group, n = 16). Rats in 24). DM group were given intraperitoneal injection of STZ (measured as 65mg/kg) in one dose. The serum Glu of DM group was measured 72 hours after the injection of STZ. The rats in DM group showed symptoms of frequent urination, drinking water and increasing food intake. Rats in NC group were given intraperitoneal administration of potassium citrate buffer solution. The rats in both groups were given general diet and observed for 7 days under the same conditions. The model group always satisfied the condition of type 1 diabetes mellitus (T1DM) model and did not return to normal in the experimental group. Rats in the latter two groups were continued to be fed with common diet. The degree of retinopathy, the concentration of TGF- 尾 1-ICAM-l in tissues and the concentration of ADPN in serum of the two groups were observed at 4 and 8 weeks, respectively. According to the experimental results and the analysis and comparison of the monitoring data, the relationship between them is discussed. Results: the level of ADPN in serum of T1DM rats was significantly lower than that of normal rats, and the lower the level of ADPN was with the prolongation of the course of disease. The expression of transforming growth factor- 尾 1 and intercellular adhesion molecule-1 in retinal tissues increased significantly with the prolongation of the course of diabetes mellitus. The severity of retinopathy was accompanied by the prolongation of the course of T1DM. Gradually aggravating. Conclusion: the serum adiponectin level of type 1 diabetic rats decreased gradually with the development of the course of disease, and the level of adiponectin decreased significantly compared with the normal group. The expression of TGF- 尾 1 and ICAM-1 in retinal tissue was significantly increased with the progression of the disease, and the degree of fibrosis was significantly aggravated. It can be concluded that with the decrease of adiponectin, the expression of TGF- 尾 1 and intercellular adhesion molecule-1 (ICAM-1) increases gradually, and the degree of retinopathy is higher.
