西格列汀对高糖下系膜细胞细胞外基质表达的影响

【摘要】:目的探讨高糖对肾小球系膜细胞细胞外基质表达的影响,以及西格列汀(SIT)对其的干预作用。方法培养肾小球系膜细胞,分为对照(Con)组、高糖(HG)组,以及高糖联合不同浓度的SIT组(HG+S1组、HG+S5组、HG+S10组和HG+S20组)。培养24 h、48 h后,用MTT法测定细胞增殖程度,ELISA检测细胞培养上清层粘连蛋白(LN)、大鼠Ⅳ型胶原蛋白(Col-Ⅳ)、转化生长因子β1(TGF-β1)蛋白表达的变化。结果高糖培养24 h、48 h后,可观察到不同浓度的SIT均可抑制高糖诱导的系膜细胞增殖;SIT组与HG组比较,差异有统计学意义;Con组肾小球系膜细胞可分泌一定量的LN、Col-Ⅳ和TGF-β1,高糖刺激后LN、Col-Ⅳ和TGF-β1分泌增加,而SIT对系膜细胞分泌物的抑制随其浓度的升高而增强。结论SIT可抑制高糖条件下系膜细胞的增殖,且能降低高糖条件下LN、Col-Ⅳ、TGF-β1的表达,SIT可能对糖尿病患者的肾脏有一定的保护作用。
