基于代谢组学对中药芪苓颗粒治疗高尿酸血症的疗效及安全性评价

【摘要】:目的:利用代谢组学方法鉴定中药芪苓颗粒治疗高尿酸血症大鼠的疗效及其安全性。方法:通过腺嘌呤灌胃及氧嗪酸钾皮下注射建立高尿酸血症模型,结合血清生化检测数据及代谢组学检测数据,对中药治疗后的安全性进行评价。结果:建立了稳定的高尿酸血症模型。建立了稳定的高尿酸血症模型,中药干预后比较,发现大鼠血清生化检测中尿酸及低密度脂蛋白水平降低,代谢组学检测中氨基酸、脂肪酸及糖类的代谢存在明显差异。结论:验证了中药芪苓颗粒治疗高尿酸血症有明确的疗效,证明其治疗过程同时对糖脂的代谢还有着一定的促进作用,且不存在代谢脏器的损伤。结果:建立了稳定的高尿酸血症模型,中药干预后比较,发现大鼠血清生化检测中尿酸及低密度脂蛋白水平降低,代谢组学检测中氨基酸、脂肪酸及糖类的代谢存在明显差异。
[Abstract]:Objective: to evaluate the efficacy and safety of Qiling granule in the treatment of hyperuricemia in rats with hyperuricemia by metabonomics. Methods: the model of hyperuricemia was established by oral administration of adenine and hypodermic injection of potassium oxazinate. The safety of traditional Chinese medicine after treatment was evaluated by combining the serum biochemical data and metabonomics data. Results: a stable hyperuricemia model was established. A stable model of hyperuricemia was established. It was found that the levels of uric acid and low density lipoprotein (LDL) in serum biochemical assay of rats were lower than those of control group. The metabolism of amino acids, fatty acids and sugars was significantly different in metabonomics. Conclusion: the curative effect of Qiling granule on hyperuricemia was verified. It was proved that Qiling granule could promote the metabolism of glucose and lipid, and there was no damage of metabolic organs in the treatment of hyperuricemia. Results: a stable model of hyperuricemia was established. After intervention, the levels of uric acid and low density lipoprotein (LDL) in serum of rats were decreased, and the metabolism of amino acids, fatty acids and sugars were significantly different in metabonomics.
