锌α2糖蛋白与高尿酸血症的相关性研究

【摘要】:目的目前锌α2糖蛋白(ZAG)与高尿酸血症(HUA)的相关性研究较少。文中旨在通过检测ZAG的mRNA、蛋白表达量在桂西地区HUA人群和尿酸(SUA)正常的健康人群之间的表达差异,初步探讨ZAG与HUA的相关性。方法收集2016年3月至2016年5月右江民族医学院附属医院HUA患者74例为HUA组,选取同期74例体检中心健康体检者作为对照组。将受试者的血液标本进行血清制备及生化指标检测、总RNA提取,采用实时荧光定量PCR检测ZAG mRNA表达量,酶联免疫吸附测定检测血清ZAG蛋白表达量,进而分析2组ZAG的mRNA和蛋白表达差异。结果 HUA组血清ZAG mRNA、ZAG蛋白表达量明显高于对照组[(13.201±3.433)vs(11.731±4.033);(6.163±1.779)vs(5.540±0.573),P0.05]。SUA与ZAG蛋白表达量(r=0.176)、BMI(r=0.161)、SBP(r=0.144)、DBP(r=0.250)、BUN(r=0.461)、CREA(r=0.649)、TG(r=0.212)呈正相关,与HDL(r=-0.333)呈负相关,均有统计学意义(P0.05)。结论 ZAG可能成为预防及治疗HUA及其相关代谢性疾病的新靶点。
[Abstract]:Objective at present, there are few studies on the correlation between zinc 伪 2 glycoprotein (ZAG) and hyperuricemia (HUA). The purpose of this study was to detect the expression of mRNA, protein in ZAG between HUA population in western Guangxi and healthy people with normal uric acid (SUA), and to explore the relationship between ZAG and HUA. Methods from March 2016 to May 2016, 74 patients with HUA in the affiliated Hospital of Youjiang Medical College for nationalities were collected as HUA group, and 74 healthy persons in physical examination center were selected as control group. The blood samples of the subjects were prepared and biochemical indexes were detected, the total RNA was extracted, the expression of ZAG mRNA was detected by real-time fluorescence quantitative PCR, and the expression of ZAG protein in serum was detected by enzyme-linked immunosorbent assay (Elisa). The difference of mRNA and protein expression between the two groups was analyzed. Results the expression of serum ZAG mRNA,ZAG protein in HUA group was significantly higher than that in control group [(13.201 卤3.433) vs (11.731 卤4.033)].
