健脾养血祛风方对脾虚型湿疹小鼠ICAM-1及VCAM-1的影响

【摘要】：目的探讨健脾养血祛风方对脾虚型湿疹小鼠模型皮损处细胞间黏附分子(ICAM-1及VCAM-1)的影响及可能的作用机制。方法取健康雄性小鼠50只,随机分为5组,即空白对照组、脾虚慢性湿疹模型组、脾虚慢性湿疹西药治疗组、脾虚慢性湿疹中药治疗高剂量组及低剂量组。中药组以健脾养血祛风方进行治疗,西药组以盐酸左西替利嗪治疗;采用免疫组化法检测小鼠耳部病变组织ICAM-1及VCAM-1的水平。结果模型组小鼠耳部病变组织ICAM-1及VCAM-1均有不同程度的阳性表达;给药后,脾虚慢性湿疹中药治疗高剂量组及西药组小鼠耳部病变组织ICAM-1和VCAM-1的表达均较模型组显著降低(P0.01)。中药高剂量组与西药组ICAM-1及VCAM-1的水平差异无统计学意义(P0.05)。结论健脾养血祛风方能够抑制脾虚型湿疹小鼠炎性细胞浸润,减轻炎症反应,其作用机制可能与抑制皮损处ICAM-1及VCAM-1的表达有关。
[Abstract]:Objective to investigate the effect and possible mechanism of invigorating spleen and nourishing blood and expelling wind on intercellular adhesion molecules (ICAM-1 and VCAM-1) in skin lesions of mice with spleen deficiency. Methods Fifty healthy male mice were randomly divided into 5 groups: blank control group, chronic eczema model group, chronic eczema treatment group, high dose group and low dose group. The traditional Chinese medicine group was treated with Jianpi Yangxue qufeng recipe, the western medicine group was treated with levcetirizine hydrochloride, and the levels of ICAM-1 and VCAM-1 in the ear lesions of mice were detected by immunohistochemical method. Results the expression of ICAM-1 and VCAM-1 in the ear lesions of the model group was significantly lower than that in the model group, and the expression of ICAM-1 and VCAM-1 in the high dose group and the western medicine group were significantly lower than that in the model group. There was no significant difference in ICAM-1 and VCAM-1 levels between high dose group and western medicine group (P 0.05). Conclusion Jianpi Yangxue qufeng recipe can inhibit the infiltration of inflammatory cells and alleviate inflammatory reaction in mice with spleen deficiency, and its mechanism may be related to the inhibition of the expression of ICAM-1 and VCAM-1 in skin lesions.
