Wnt10b对体外培养小鼠触须毛囊生长的作用研究

【摘要】:目的:研究Wnt10b对体外培养小鼠触须毛囊生长的作用。方法:构建Dct-Lac Z小鼠触须毛囊体外培养模型。用腺病毒Ad-Wnt10b和Ad-GFP感染触须毛囊2 d、4 d并对触须生长长度进行测量。X-gal染色检测触须毛囊中黑素细胞数量。Western blot检测酪氨酸相关蛋白1(tyrosinase-related protein 1,TRP-1)、酪氨酸酶(tyrosinase,TYR)蛋白的表达水平。结果:腺病毒感染4 d后长度测量结果显示Wnt10b处理组的触须毛囊生长长度明显高于GFP对照组(0.58±0.03 vs.0.09±0.05,P=0.000);X-gal染色结果显示Wnt10b处理组的黑素细胞数量明显多于GFP对照组;Western blot结果显示Wnt10b处理组的触须毛囊中TRP-1和TYR的蛋白表达明显升高(1.14±0.16 vs.0.55±0.22,1.34±0.33 vs.0.77±0.09;P=0.020,P=0.045)。结论:Wnt10b能够促进小鼠触须毛囊的体外生长。
[Abstract]:Objective: to study the effect of Wnt10b on the growth of mouse hair follicles in vitro. Methods: Dct-Lac Z mouse hair follicles were cultured in vitro. The hair follicles were infected with adenovirus Ad-Wnt10b and Ad-GFP for 2 d and the length of the hair follicles was measured. The number of melanocytes in the hair follicles was detected by X-gal staining. The expression of tyrosine associated protein 1 (tyrosinase-related protein 1) and tyrosinase (tyrosinase,TYR) protein in the hair follicles was detected by. Western blot. Results: the length measurement after 4 days of adenovirus infection showed that the growth length of hair follicles in Wnt10b treated group was significantly longer than that in GFP control group (0.58 卤0.03 卤0.05 vs.0.09 卤0.05 P0. 000), and X-gal staining showed that the number of melanocytes in Wnt10b treated group was significantly larger than that in GFP control group. The expression of TRP-1 and TYR in the hair follicles of the Wnt10b treated group was significantly increased (1.14 卤0.16 vs.0.55 卤0.22 vs.0.55 卤0.33 vs.0.77 卤0.33 vs.0.77 卤0.09 vs.0.77 卤0.020 TYR 0.045). Conclusion: Wnt10b can promote the growth of mouse hair follicles in vitro.
