早期脑出血后细胞凋亡与p75NTR、TrkA的相关性研究

【摘要】:目的:探讨p75NTR、TrkA在早期人高血压脑出血后的表达,并初步阐明与细胞凋亡的联系。 方法:收集脑出血后血肿周围不同部位组织标本;取血肿表面组为A组,半暗带组为B组,脑皮质区组为C组,其中,A组、B组为实验组,C组为对照组;并在A、B、C三组中将标本按出血时间(7h、≤7h)、出血量(60ml、60ml)分组。采用免疫组化检测p75NTR、TrkA在早期人脑出血距离血肿不同区域中的表达; Tunel方法检测出血后各部位的凋亡细胞率;统计学方法分析并阐明p75NTR、TrkA及凋亡细胞在时空上的关系。 结果: 1、 Tunel法检测脑出血后A组凋亡细胞B组C组;统计学分析: A36.30%±2.83%,B22.55%±3.49%,C17.70%±2.62%,,p0.01。 2、在早期人脑出血中越靠近血肿表面p75NTR表达增加,A组p75NTR阳性细胞率B组C组(х2=15.501v=2p0.01);各组p75NTR表达与凋亡细胞率比较变化程度一致,呈正比(A组:r=0.6396,p=0.0024;B组:r=0.7889,p0.0001;C组:r=0.8772,p0.0001);TrkA在各组中有表达,但三组标本中TrkA无差异(х2=0.877v=2p0.05),与细胞凋亡无明显联系;p75NTR/TrkA比率在A组中高于其余二组(F=4.851,p=0.011)。 3、p75NTR在7h组中表达明显强于≤7h组(Z=-2.566p=0.01),在60ml组中表达亦强于60ml组(Z=-2.692p=0.007)。而TrkA在上述组中无表达差异(p0.05)。 结论: 1、HICH后脑组织中凋亡细胞由远及近增多; 2、脑出血后p75NTR在TrkA低表达或者无活性的环境中引发细胞凋亡;越靠近血肿表面,其促凋亡作用越强; 3、TrkA在早期HICH脑组织中无表达差异,与细胞凋亡无相关; 4、p75NTR/TrkA比率的变化影响出血后细胞凋亡。
[Abstract]:Aim: to investigate the expression of p75 NTRG TrkA after early hypertensive intracerebral hemorrhage, and to elucidate the relationship between p75 NTR TrkA and apoptosis.
