恩再适对CCI大鼠痛觉过敏及脊髓NR2B、NF-κB和iNOS表达的影响

【摘要】:目的:1.观察坐骨神经慢性损伤大鼠机械痛阈及热痛阈的变化,测定坐骨神经慢性压迫性损伤(Chronic constriction injury, CCI)大鼠脊髓N-甲基D-天冬氨酸受体2B亚基(N-methyl-D-aspartic acid receptor2B, NR2B)、核因子-κB(nuclear factor-kappa B, NF-κB)和诱导型一氧化氮合酶(Inducible nitric oxide synthase, iNOS)的表达,探讨神经病理性疼痛的发病机制。 2.观察恩再适对CCI大鼠痛觉过敏及脊髓NR2B、NF-κB和iNOS表达的影响。 方法:1.56只180-220g雄性SD大鼠随机分为2组:分别为Sham组(假手术组,n=8)和CCI组(CCI组,n=48)。于术前1d、术后1d、4d、7d、14d、21d测定机械痛阈(mechanical withdrawal threshold, MWT)和热痛阈(paw withdrawal latency, PWL)后处死,取L4-6脊髓,采用逆转录-聚合酶链式反应(Reverse transcription-Polymerase chain reaction, RT-PCR)和蛋白质印迹法(Western blot, WB)检测脊髓NR2B、NF-κB和iNOS的表达。 2.96只180-220g雄性SD大鼠随机分为2组:分别CCI组(CCI模型组,n=48)和A组(恩再适封闭治疗组,n=48)。测定MWT和PWL后处死大鼠,取L4-6脊髓,采用实时定量PCR(Real time quantitative PCR, qPCR)和WB方法检测脊髓NR2B、NF-κB和iNOS的表达。 结果:1.与Sham组相比,CCI组术侧从术后1d开始出现MWT值下降(P0.01,n=8),术后7d降至最低(P0.01,n=8)。术后14d时仍处于较低水平(P0.01,n=8)。与Sham组相比,CCI组术侧从术后1d开始出现PWL值下降(P0.01,n=8),术后7d降至最低(P0.01,n=8)。术后14d时仍处于较低水平(P0.01,,n=8)。脊髓组织中NR2B和NF-κB于损伤后1d开始增加(P0.01,n=4);7d达到高峰,与术前相比有显著性意义(P0.01,n=4)。术后14d虽有所降低,但仍维持在较高水平(P0.01,n=4)。iNOS mRNA的表达于术后1d开始增加(P0.01,n=4),术后14d达到高峰,显著高于术前(P0.01,n=4),术后21d时仍保持较高水平表达(P0.01,n=4)。 iNOS mRNA与NR2BmRNA和NF-κB mRNA的表达呈显著正相关(r=0.833,P0.05;r=0.842,P0.05)。 2.术后CCI组MWT及PWL较术前明显降低(n=8,P0.01),A组MWT及PWL明显高于CCI组(n=8, P0.05)。 CCI组术后脊髓NR2B、NF-κB和iNOS的表达较术前明显升高(n=4,P0.05),A组脊髓NR2B、NF-κB和iNOS的表达较CCI组明显降低(n=4,P0.05)。 结论:1.CCI术后大鼠脊髓中NR2B、NF-κB及下游iNOS表达增加,可能与神经病理性疼痛的产生和维持有关。 2.恩再适封闭治疗能够改善CCI大鼠痛觉过敏,其作用效应与影响脊髓NR2B、NF-κB和iNOS的表达有关。
[Abstract]:Purpose 1.To observe the changes of mechanical pain threshold and thermal pain threshold in rats with chronic sciatic nerve injury.The expression of N-methyl-D-aspartic acid receptor 2B subunit N-methyl-D-aspartic acid receptor 2B, nuclear factor- 魏 B(nuclear factor-kappa B (NF- 魏 B) and inducible nitric oxide synthase (iNOS) in the spinal cord of rats with chronic compression injury of sciatic nerve was determined.To explore the pathogenesis of neuropathic pain.2.To observe the effects of enrezol on hyperalgesia and the expression of NF- 魏 B and iNOS in spinal cord of CCI rats.Methods 1.56 male Sprague-Dawley rats (180-220 g) were randomly divided into two groups: Sham group (sham-operated group) and CCI group.The mechanical withdrawal threshold (MWT) and the thermal pain threshold of paw withdrawal (PWL) were measured 1 day before operation and 4 days after operation. The spinal cord L4-6 was killed. Reverse transcription-Polymerase chain reactionation (RT-PCR) and Western blotting were used to detect the expression of NR2BN-NF- 魏 B and iNOS in spinal cord.2.96 male Sprague-Dawley rats (180-220 g) were randomly divided into two groups: CCI group (CCI model group) and group A (group A).MWT and PWL were used to determine the expression of NF- 魏 B and iNOS in spinal cord NR2BN- 魏 B, and the L4-6 spinal cord was harvested. PCR(Real time quantitative PCR (qPCR) and WB were used to detect the expression of NF- 魏 B and iNOS in spinal cord.The result is 1: 1.Compared with the Sham group, the MWT value decreased from the first day after operation to the lowest value at the 7th day after operation.On the 14th day after operation, it was still at a lower level (P 0.01).Compared with the Sham group, the PWL value decreased from the first day after operation to the lowest value at the 7th day after operation.On the 14th day after operation, it was still at a lower level (P 0.01).The levels of NR2B and NF- 魏 B in spinal cord tissue began to increase at 1 day after injury and reached the peak at 7 days after injury, which was significantly higher than that before operation.On the 14th day after operation, the expression of P0. 01 mRNA was maintained at a high level, but the expression of iNOS mRNA increased on the 1st day after operation, and reached the peak on the 14th day after operation.The expression of iNOS mRNA was significantly higher than that of preoperation (P 0.01), and remained high at 21 days after operation. There was a significant positive correlation between the expression of iNOS mRNA and NR2BmRNA and NF- 魏 B mRNA.2.The MWT and PWL in CCI group were significantly lower than those in pre-operation group (P 0.01). The MWT and PWL in group A were significantly higher than those in group CCI (P 0.05).The expression of NF- 魏 B and iNOS in spinal cord of CCI group was significantly higher than that before operation. The expression of NF- 魏 B and iNOS in group A was significantly lower than that in group CCI.Conclusion 1. The increased expression of NF- 魏 B and downstream iNOS in spinal cord of rats after CCI may be related to the generation and maintenance of neuropathic pain.2.Enreclosure block therapy can improve hyperalgesia in CCI rats, and its effect is related to the effect on the expression of NF- 魏 B and iNOS in spinal cord NR2BU.
