EphA2在脑膜瘤中的表达及意义

【摘要】：目的：通过检测脑膜瘤组织中促红细胞生成素肝细胞(erythropoietin-producing hepatocellular Eph)受体酪氨酸激酶A2的表达情况，从而探索其与脑膜瘤发生、分级及瘤周水肿的关系。方法：①脑膜瘤标本共60例，均取自泸州医学院附属医院神经外科2012年1月至2013年1月手术切除病理标本。所选病例均为原发首次手术病例，术前未行药物及放射治疗，各脑膜瘤标本的病理资料和临床资料（包括影像资料）均完整。对照组25例，为内减压术后所取正常蛛网膜组织。②脑膜瘤病理标本按世界卫生组织（WHO）2007年标准对其进行病理分级并分为两组。③所有的对照组标本均在分离人体后12小时之内进行冷冻、固定处理。④对脑膜瘤及正常脑膜组织进行免疫组化链霉菌抗生物素蛋白-过氧化物酶连结（streptavidin-perosidase S-P）法染色处理，检测其中的EphA2表达情况。⑤观察并记录结果。结果：①正常脑膜中，EphA2表达阴性17例，阳性8例；在脑膜瘤组织中阴性12例，阳性48例，两者之间差异有显著性（2=18.089，P=0.0000.05)。②Ⅰ级脑膜瘤组织中EphA2阴性10例，阳性41例；在Ⅱ级脑膜瘤组织中，EphA2阴性2例，阳性7例，Ⅰ级和Ⅱ级脑膜瘤组织中EphA2阳性表达率差异无显著性（2=0.033，P=0.58＞0.05）。③Ⅰ级脑膜瘤伴有瘤周水肿的有21例，水肿率41.2%，Ⅱ级脑膜瘤伴有瘤周水肿的有8例，水肿率88.9%，差异有显著性（2=6.974，P=0.010.05）。④在脑膜瘤中，EphA2阳性的瘤周水肿率与阴性的瘤周水肿率差异无显著性（2=1.352，P=0.201＞0.05）。结论：①脑膜瘤组织中EphA2呈过度表达，，提示EphA2可能参与了脑膜瘤的发生发展。②本实验结果提示EphA2与脑膜瘤的分级无关，原因可能与实验中Ⅱ级脑膜瘤总例数少有关，有待我们做进一步的研究。③Ⅱ级脑膜瘤较Ⅰ级脑膜瘤更易发生瘤周水肿。④本实验结果提示EphA2与脑膜瘤瘤周水肿无关，可能同样与实验中Ⅱ级脑膜瘤总例数少有关。
[Abstract]:Aim: to investigate the expression of erythropoietin (erythropoietin-producing hepatocellular Eph) receptor tyrosine kinase A2 in meningiomas and its relationship with meningioma occurrence, grading and peritumoral edema. Methods: 1 A total of 60 meningioma specimens were obtained from neurosurgery department of affiliated Hospital of Luzhou Medical College from January 2012 to January 2013. All the selected cases were primary operation cases. No drug and radiotherapy were performed before operation. The pathological and clinical data (including imaging data) of all meningioma specimens were complete. Control group 25 cases, Normal arachnoid tissue was taken after internal decompression. 2 pathological specimens of meningioma were classified according to WHO (WHO) 2007 standard and divided into two groups. Frozen within hours, 4 Immunohistochemical staining of streptomycin and peroxidase (streptavidin-perosidase S-P) was used to detect the expression of EphA2 in meningiomas and normal meningeal tissues. 5 the results were observed and recorded. Results: 1 in normal meninges, 17 cases were negative in EphA2 expression, 8 cases were positive; In meningioma tissue, 12 cases were negative, 48 cases were positive, there was significant difference between the two groups. 2 in grade 鈪
