低能量激光照射对体外培养嗅鞘细胞的影响

【摘要】: 目的:研究低能量半导体激光对体外培养嗅粘膜嗅鞘细胞增殖的影响。方法:取成年SD大鼠嗅粘膜,采用差速贴壁法获得嗅鞘细胞,对细胞进行不同能量密度的激光照射(在10.3mW /cm2功率密度下分别照射30秒、60秒、120秒,每日一次,连续三天),采用细胞计数法和MTT法分别检测照射后不同时间细胞增殖的变化。结果:培养3天时,不同时间照射组和对照组细胞增殖无差异。培养5天和8天时,不同时间照射组细胞数均高于对照组(p0.05),有统计学意义;60秒和120秒照射组OD值高于对照组(p0.05),有统计学意义;但30秒照射组OD值与对照组无差异。其中以60秒照射组细胞增值最为明显。结论:低能量激光对嗅粘膜嗅鞘细胞的增殖有促进作用并且与照射的能量密度有关。最佳照射时间是每日一次,连续三天。
[Abstract]:Aim: to study the effect of low energy semiconductor laser on the proliferation of olfactory ensheathing cells in vitro. Methods: olfactory ensheathing cells were obtained from olfactory mucosa of adult Sprague-Dawley rats. The cells were irradiated with laser with different energy density (30 seconds, 60 seconds, 120 seconds, once a day, respectively) at 10.3mW / cm2 power density. Cell count method and MTT method were used to detect cell proliferation at different time after irradiation for three consecutive days. Results: there was no difference in cell proliferation between irradiation group and control group at 3 days. At 5 and 8 days, the number of cells in the irradiation group was higher than that in the control group (P 0.05), and the OD value in the irradiation group at 60 seconds and 120 seconds was higher than that in the control group (P 0.05), but there was no difference in OD value between the 30 second group and the control group. In the 60 second group, the cell proliferation was the most obvious. Conclusion: low energy laser can promote the proliferation of olfactory ensheathing cells of olfactory mucosa and is related to the energy density of irradiation. The best exposure time is once a day for three consecutive days.
