重组双歧杆菌-LTB疫苗免疫大鼠后抗体分泌部位的组织细胞定位

【摘要】: 双歧杆菌(Bifidobacterium)是人类肠道内重要的益生菌,对人体具有很好的保健功能。基于双歧杆菌表达系统的重组疫苗对人体可以产生良好的保护作用,既能使人体产生抵抗病原菌的抗体,又可以调整肠道内菌群的平衡。因此我们选用双歧杆菌作为重组疫苗的受体菌。 产肠毒素大肠杆菌(ETEC)是引起腹泻的主要致病菌。它导致数以万计的人们尤其是发展中国家的婴幼儿腹泻,同时它也是旅行者腹泻的最常见病原菌。因此,一个安全有效的抵抗ETEC腹泻的疫苗对公众健康是非常重要的。 目的:为了进一步研究重组双歧杆菌-LTB疫苗免疫于机体后引发的免疫反应,对免疫大鼠的抗体分泌部位进行组织细胞定位。 方法:通过ELISA方法,间接的证明了机体产生了IgG抗体和IgA抗体;而通过免疫组化方法,对产生sIgA的组织细胞进行进一步的定位。 结果:免疫后的SD大鼠血清中产生了特异性的抗体IgG,肠道内产生了特异性分泌型抗体sIgA。免疫组化的图片显示抗体sIgA主要是由小肠组织产生的,分泌的细胞为小肠绒毛固有层细胞和小肠腺细胞。随着免疫次数的增加,抗体分泌量明显增加,疫苗激发的免疫反应使机体大量分泌sIgA。 结论:重组双歧杆菌-LTB疫苗免疫大鼠后抗体sIgA分泌组织为小肠,分泌细胞为小肠绒毛固有层细胞和小肠腺细胞。
[Abstract]:Bifidobacterium (Bifidobacterium) is an important probiotic in human intestinal tract and has good health function for human body. The recombinant vaccine based on Bifidobacterium expression system can produce a good protective effect on human body, which can make the body produce antibodies against pathogenic bacteria and adjust the balance of bacteria in the intestinal tract. So we choose two pairs. Sod is the recipient of the recombinant vaccine.
Enterotoxigenic Escherichia coli (ETEC) is a major pathogen causing diarrhea. It causes tens of thousands of people, especially in developing countries, and is also the most common pathogen of traveller diarrhoea. Therefore, a safe and effective vaccine against ETEC diarrhea is very important to public health.
Objective: in order to further study the immune response induced by the recombinant Bifidobacterium -LTB Vaccine Immunized by the body, the tissue cell location of the antibody secreting parts of the immune rats was carried out.
Methods: the ELISA method was used to indirectly prove that the body produced IgG and IgA antibodies, and the tissue cells that produced sIgA were further localized by immunohistochemical method.
Results: the specific antibody IgG was produced in the serum of SD rats after immunization, and a specific secretory antibody sIgA. was produced in the intestinal tract. The antibody sIgA was produced mainly by small intestinal tissue, and the secreted cells were small intestinal villus propria cells and small intestinal gland cells. The immune response stimulated by the vaccine increases the secretion of sIgA. by the body.
Conclusion: after immunization with recombinant Bifidobacterium -LTB vaccine, the sIgA secreted tissue was small intestine, and secretory cells were small intestinal villus lamina propria cells and small intestine gland cells.
