瓜蒌皮干预急性心肌梗死的药效与机制研究

【摘要】:目的:以急性心肌梗死模型大鼠为研究对象,通过研究瓜蒌皮注射液对该模型大鼠血浆中肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、谷草转氨酶(AST)、α-羟丁酸脱氢酶(α-HBDH)5项心肌酶的活性,内皮素(ET)、内皮型一氧化氮合酶(eNOS)、一氧化氮(NO)、6-酮-前列环素(6-Keto-PGF1α)、血栓素B2(TXB2)的含量,心肌中丙二醛(MDA)和超氧化物歧化酶(SOD)活性的影响,探讨瓜蒌皮干预急性心肌梗死损伤的作用效果,为瓜蒌皮的临床应用提供实验依据。 方法:通过结扎大鼠左冠状动脉前降支,制备大鼠急性心肌梗死模型。用水合氯醛麻醉,,取血、心肌,心肌进行切片,以氯化三苯四氮唑(TTC)染色来测定心肌梗死率;采用半自动生化分析仪测定血浆中CK等5项心肌酶活性,以确定缺血心肌坏死程度;生化试剂盒检测心肌组织中MDA及SOD活性,探讨药物清除大鼠心肌氧自由基能力;并采用ELISA法检测NO、eNOS、ET、6-ketone-PGF1α、TXB2等指标,探讨药物对血管内皮的保护作用和预防血栓形成的效果。 结果:与模型组相比,瓜蒌皮组可显著降低心肌梗死大鼠心肌梗死率,维护缺血心肌正常生理功能,明显降低CK、CK-MB、LDH、AST、α-HBDH5项心肌酶活性,升高心肌组织SOD活性,降低MDA含量,增加血液中NO、eNOS、6-ketone-PGF1α的含量,减少ET、TXB2的含量。 结论:瓜蒌皮对大鼠急性心肌梗死有明显保护作用,其机制可能与抑制缺血心肌脂质过氧化,提高组织抵抗氧自由基损伤能力,促进血管舒张,抑制血管收缩,抑制血小板的活化、聚集,减小血栓形成等有关。
[Abstract]:Objective: to study the acute myocardial infarction model in rats. The effects of trichosanthes peel injection on the activities of creatine kinase (CK), creatine kinase isoenzyme (CK-MBN), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), 伪 -hydroxybutyrate dehydrogenase (伪 -HBDHH) in plasma of the model rats were studied. The contents of endothelin (et), endothelium-nitric oxide synthase (Enos), 6-Keto-PGF1 伪 (6-Keto-PGF1 伪), thromboxane B2TXB2 (TXB2), the activities of malondialdehyde (MDA) and superoxide dismutase (SOD) in myocardium, and the effects of trichosanthes on acute myocardial infarction injury were investigated. Methods: rat acute myocardial infarction model was established by ligating the anterior descending branch of left coronary artery. The myocardial infarction rate was determined by using chloral hydrate anesthesia, blood and myocardial sections, and the myocardial enzymes activity of CK and other 5 items in plasma were measured by semi-automatic biochemical analyzer, and the myocardial infarction rate was determined by TTC staining. In order to determine the degree of ischemic myocardial necrosis, the activity of MDA and SOD in myocardial tissue was detected by biochemical kit, and the ability of drug scavenging oxygen free radical in myocardium of rats was studied. To investigate the protective effect of drugs on vascular endothelium and the effect of preventing thrombosis. Results: compared with model group, trichosanthes skin group could significantly reduce myocardial infarction rate and maintain normal physiological function of ischemic myocardium in rats with myocardial infarction. The activity of myocardial enzymes of CK CK-MBN LDHN AST, 伪 -HBDH5 was significantly decreased, the activity of SOD was increased, the content of MDA was decreased, the content of NON-NOSS-6-ketone-PGF 1 伪 in blood was increased, and the content of ETT XB2 was decreased. Conclusion: Trichosanthes peel has obvious protective effect on acute myocardial infarction in rats. The mechanism may be related to inhibition of lipid peroxidation in ischemic myocardium, enhancement of tissue resistance to oxygen free radical damage, promotion of vasodilation, inhibition of vasoconstriction, inhibition of platelet activation, aggregation and reduction of thrombosis.
