莪术对局灶性脑缺血模型大鼠的神经保护作用

【摘要】:目的:探讨莪术对局灶性脑缺血模型大鼠的神经保护作用及机制。方法:健康成年雄性SD大鼠60只,随机分为6组,分别为假手术对照组、模型对照组、尼莫地平组、莪术高、中、低剂量组。除了假手术对照组,其余组大鼠采用线栓法制作大鼠大脑中动脉梗阻模型。造模前预防给药7d,造模后给药3d,每天1次。各组大鼠进行神经行为学评分,断头取脑进行TTC染色,检测大鼠血清中乳酸脱氢酶(LDH)、肌酸激酶(CK)、谷氨酸(Glu)含量以及脑含水量、脑梗死体积百分比改变。结果:与模型对照组相比,莪术高剂量组LDH、CK、Glu含量以及脑含水量、脑梗死体积百分比有明显降低(P0.05),莪术中剂量组LDH、Glu含量和脑梗死体积百分比也有明显降低(P0.05),而莪术中剂量组CK、脑含水量和莪术低剂量组LDH、CK、Glu、脑含水量和脑梗死体积百分比与模型对照组相比差异无统计学意义(P0.05)。说明莪术在一定剂量下能有效地降低局灶性脑缺血大鼠LDH、CK和Glu含量以及脑含水量、脑梗死体积百分比。结论:莪术对局灶性脑缺血模型大鼠具有一定的神经保护作用,这种作用可能与保护缺血区脑组织、减轻脑水肿、抗氧自由基,减少兴奋性氨基酸毒性有关。
[Abstract]:Aim: to investigate the neuroprotective effect and mechanism of Zedoary on focal cerebral ischemia in rats. Methods: sixty adult male SD rats were randomly divided into six groups: sham operation control group, model control group, nimodipine group, zedoary high, middle and low dose groups. In addition to the sham operation control group, the middle cerebral artery obstruction model was made by the method of thread embolization. Prophylactic administration was given for 7 days before modeling, and once a day for 3 days after modeling. The neurobehavioral score and TTC staining were used to detect the contents of lactate dehydrogenase (LDH), creatine kinase (CK), glutamate (Glu), brain water content and the percentage of cerebral infarction volume. Results: compared with the model control group, the content of Glu and the water content of brain in the high dose group of Rhizoma Curcumae were higher than those in the control group. The percentage of cerebral infarction volume decreased significantly (P 0.05), and the content of LDH Glu and the percentage of cerebral infarction volume decreased significantly in the middle dose group of zedoary curcuma, while in the middle dose group of zedoary, the contents of CK, the water content in the brain and LDHK CK Glul in the low dose group of zedoary, the content of water in the brain and the volume of cerebral infarction in the middle dose group were 100. There was no significant difference between the score ratio and the model control group (P 0.05). The results showed that zedoary curcuma could effectively reduce the contents of LDHCK and Glu, the water content of brain and the percentage of cerebral infarction volume in rats with focal cerebral ischemia at a certain dose. Conclusion: zedoary has a certain neuroprotective effect on focal cerebral ischemia rats, which may be related to the protection of ischemic brain tissue, the reduction of brain edema, anti-oxygen free radical, and the reduction of excitatory amino acid toxicity.
