大鼠自然衰老过程中脊髓突触体及血小板膜外周型苯二氮卓受体的变化

【摘要】: 目的:应用放射性配基结合试验,探讨自然衰老过程中脊髓突触体外周型苯二氮卓受体(PBRs)的动态变化以及与外周血血小板膜PBRs的相关性。 方法:实验动物SD大鼠(雌雄各半)分为3月龄和24月龄2组。动物断头处死后,迅速取脊髓。采用梯度离心技术制备脊髓突触体,低渗溶血法制备外周血血小板膜。应用放射配基[~3H]PK11195结合实验测定PBRs结合活力。 结果:各组动物雄性、雌性间比较脊髓突触体、外周血血小板膜PBRs结合活性差异均无显著意义(P>0.05)。随着鼠龄增加,脊髓突触体及外周血血小板膜[~3H]PK11195结合活性显著下降。外周血血小板膜[~3H]PK11195结合活性与脊髓突触体[~3H]PK11195结合活性间具有显著相关性(r=0.985,P<0.001)。 结论:脊髓突触体PBRs水平呈增龄下降改变,外周血血小板膜[~3H]PK11195结合活性能够反映脊髓组织该指标的变化。
[Abstract]:Aim: to investigate the dynamic changes of peripheral benzodiazepine receptor (PBRs) in spinal cord synapses during natural senescence and the correlation between PBRs and PBRs in peripheral blood platelets. Methods: Sprague-Dawley rats (male and female) were divided into 2 groups: 3 months old and 24 months old. After the animals were killed, the spinal cord was quickly removed. Spinal synaptosomes were prepared by gradient centrifugation and platelet membranes were prepared by hypotonic hemolysis. The binding activity of PBRs was determined by radioligand [3 H] PK11195 binding assay. Results: there was no significant difference in PBRs binding activity between male and female spinal cord synaptosomes and peripheral blood platelets (P > 0.05). The binding activity of PK11195 in synaptosomes of spinal cord and platelets in peripheral blood decreased significantly with aging. There was a significant correlation between the binding activity of PK11195 in peripheral blood platelet membrane and the binding activity of PK11195 in spinal synaptosomes (r = 0.985, P < 0.001). Conclusion: the level of PBRs in spinal synaptosomes decreased with age, and the binding activity of [3H] PK11195 in peripheral blood platelets could reflect the changes in spinal cord tissues.
