溶血磷脂酸影响胚胎神经干细胞增殖和分化的初步观察

神经干细胞(neural stem cells,NSCs)广泛存在于胚胎脑组织中,也存在于成年哺乳动物的神经系统。NSCs具有自我更新能力,能够定向分化为神经元、星形胶质细胞和少突胶质细胞。NSCs的这一特点使其有望成为体外移植治疗神经系统损伤和退行性疾病的重要材料。 溶血磷脂酸(lysophospatidic acid,LPA)是一种简单的磷脂类物质,是血清的正常成份,而且正常情况下也存在于脑组织内。LPA的受体,LPAl-3,在多种细胞的细胞膜上有不同表达。LPA与G蛋白耦联的受体结合后,激活下游信号传导通路,从而调节体内各种细胞,包括神经元、神经胶质细胞等的功能活动。LPA在细胞水平的作用很广泛,主要是影响细胞形态,促进细胞增殖和存活,诱导细胞凋亡,改变离子电流和钙胞内释放等。Contos等曾报道,1.0 μmo/L LPA可以促进小鼠胚胎NSCs的增殖。但也有文献报道LPA的作用具有两面性,即不同浓度的LPA对同一细胞的功能影响不同;而且,LPA对来自大鼠胚胎的NSCs的增殖和分化方面的影响如何,并未见有关报道。因此,本实验第一部分探讨了LPA对体...
